贯筋藤水提物对小鼠的镇静催眠作用 被引量:1

摘要 【目的】探究贯筋藤水提物对小鼠镇静、催眠及抗惊厥作用的影响.【方法】首先利用冷凝回流小提法制备贯筋藤水提物;将ICR小鼠按体质量随机分为贯筋藤水提物(高、中、低)剂量组、艾司唑仑片组及空白组,采用光电计数法检测贯筋藤水提物对小鼠自发活动的影响,利用戊巴比妥钠阈下催眠剂量和延长戊巴比妥钠睡眠时间试验,观察并记录贯筋藤水提物对小鼠催眠的作用,同时利用一些致惊厥药物如戊四唑、印防己毒素及士的宁考察了贯筋藤水提物的抗惊厥作用.【结果】与空白组比较,贯筋藤水提物各剂量组、艾司唑仑片组能显著减少小鼠的自主活动次数(P<0.05);明显增加戊巴比妥钠阈下剂量小鼠睡眠个数(P<0.05);显著缩短阈剂量戊巴比妥钠诱导小鼠睡眠潜伏期,延长睡眠持续时间(P<0.05);表现出一定的抗惊厥作用,明显降低小鼠死亡率(P<0.05).【结论】贯筋藤水提物具有很好的镇静催眠及抗惊厥作用. 【Objective】To determine the sedative-hypnotic and anti-seizure effects of the water extract of Dregea sinensis Hemsl(WEDSH)in mice.【Method】Firstly,we utilized water condensation reflux to prepare the crude extraction during the experiment.Secondly,according to the weight,the ICR mice were randomly divided into five groups,including different dose of WEDSH group,estazolam group and control group.Thirdly,using the machine,which can observe the spontaneous activity of mice and the hypnotic effect of the WEDSH was assessed by pentobarbital sodium induced sleeping test.Finally,the anti-convulsant activitiy of the WEDSH was evaluated by the pentylenetetrazole,picrotoxin and strychnine induced convulsion animal model.【Result】Compared with the control group,the results indicated that the high,middle and low doses of the WEDSH and estazolam were dramatically inhibited spontaneous activities of the mice(P<0.05);to the pentobarbital-induced sleeping,the number of mice falling asleep in sub-threshold dose of pentobarbital(30 mg/kg)was obviously increased(P<0.05),the sleep onset was shortened significantly and the sleep duration was dramatically prolonged(P<0.05)in threshold dose of pentobarbital-induced(50 mg/kg)mice;different dose of the WEDSH showed anti-seizure effects on mice and also significantly reduced the mortality(P<0.05).【Conclusion】Consequently,the WEDSH has good sedative-hypnotic and anti-seizure effects in mice with dose-dependently relationship.
