摘要
目的研究血清微小RNA miR-32-5p、miR-409-3p在宫颈癌诊断和预后中的临床价值。方法选择2015年1月至2016年12月在该院行宫颈癌手术切除的患者112例为宫颈癌组,选择同期在该院就诊确诊为子宫颈上皮内瘤变(CIN)患者72例为CIN组,选择体检健康者45例为健康对照组。观察各组血清miR-32-5p、miR-409-3p、人绒毛膜促性腺激素(β-HCG)、血清糖类抗原(CA)125水平的变化,并分析血清miR-32-5p、miR-409-3p表达与临床指标、诊断和预后的关系。结果宫颈癌组术前血清miR-32-5p、β-HCG水平明显高于CIN组和健康对照组(P<0.05),CIN组明显高于健康对照组(P<0.05),宫颈癌组术后血清miR-32-5p、β-HCG水平较术前明显降低(P<0.05)。宫颈癌组的术前血清miR-409-3p水平明显低于CIN组和健康对照组(P<0.05),CIN组明显低于健康对照组(P<0.05),宫颈癌组术后血清miR-409-3p水平较术前明显升高(P<0.05)。生存组血清miR-32-5p水平明显低于死亡组(P<0.05),而血清miR-409-3p水平明显高于死亡组(P<0.05)。血清miR-32-5p、miR-409-3p水平诊断宫颈癌的效能明显优于β-HCG,miR-32-5p和miR-409-3p联合检测诊断和预测3年内出现死亡的效能明显优于单项指标。结论miR-32-5p、miR-409-3p参与宫颈癌的发生、发展,在宫颈癌的诊断和预后判断中具有重要的临床意义。
Objective To study the clinical value of serum microRNA miR-32-5p and miR-409-3p in the diagnosis and prognosis of cervical cancer.Methods From January 2015 to December 2016,112 patients who underwent cervical cancer resection in the hospital were selected as cervical cancer group,72 patients with cervical intraepithelial neoplasia(CIN)diagnosed in the hospital during the same period were selected as CIN group,and 45 healthy people were selected as healthy control group.The levels of serum miR-32-5p,miR-409-3p,human chorionic gonadotropin(β-HCG)and serum carbohydrate antigen(CA)125 were observed,and the relationship between serum miR-32-5p,miR-409-3p and clinical indicators,diagnosis and prognosis was observed.Results The preaperative serum levels of miR-32-5p andβ-HCG in cervical cancer group were significantly higher than those in CIN group and healthy control group(P<0.05),and those in CIN group were significantly higher than those in healthy control group(P<0.05),and serum levels after operation were significantly lower than those before operation(P<0.05).The preaperative serum level of miR-409-3p in cervical cancer group was significantly lower than that in CIN group and healthy control group(P<0.05),and that in CIN group was significantly lower than that in healthy control group(P<0.05),and the serum level after operation was significantly higher than that before operation(P<0.05).The serum level of miR-32-5p in the survival group was significantly lower than that in the death group(P<0.05),while the serum miR-409-3p level was significantly higher than that in the death group(P<0.05).The serum levels of miR-32-5p and miR-409-3p were significantly better thanβ-HCG in the diagnosis of cervical cancer.The combined detection of miR-32-5p and miR-409-3p was superior to single index in the diagnosis and prediction of death within 3 years.Conclusion miR-32-5p and miR-409-3p are involved in the occurrence and development of cervical cancer,and have important clinical significance in the diagnosis and prognosis of cervical cancer.
