非器质性厌食症儿童肠道菌群的多样性分析 被引量:5

摘要 目的对厌食症儿童肠道菌群的多样性进行研究,以期为临床合理选择微生态制剂提供一定的理论依据。方法将2018年5-10月在该院儿童保健和儿科门诊就诊的24例厌食症儿童设为厌食症组,25例健康体检儿童设为健康对照组。采集两组儿童大便标本,经聚合酶链反应、高通量测序后进行生物信息学分析。结果在门分类水平,厌食症组儿童大便标本中检测出14个物种,健康对照组儿童大便标本中检测出11个物种;在属分类水平,厌食症组儿童大便标本中检测出193个物种,健康对照组儿童大便标本中检测出180个物种;在种分类水平,厌食症组儿童大便标本中检测出364个物种,健康对照组儿童大便标本中检测出326个物种。两组儿童大便标本检出细菌在门分类水平上的优势菌相同,分别为厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)、放线菌门(Actinobacteria)、变形菌门(Proteobacteria)和疣微菌门(Verrucomicrobia),在属分类水平上均有17种优势菌。两组儿童大便标本检出细菌在门分类水平上Firmicutes、Proteobacteria和Verrucomicrobia组成比较,差异均无统计学意义(P>0.05);厌食症组儿童大便标本检出细菌中Bacteroidetes含量大于健康对照组,Actinobacteria含量小于健康对照组。两组儿童大便标本检出细菌在属分类水平中的13个优势菌属比较,差异均有统计学意义(P<0.05),厌食症组儿童大便标本检出细菌中双歧杆菌属(Bifidobacterium)、布劳特氏菌属(Blautia)、链球菌属(Escherichia-Shigella)、Erysipelatoclostridium、爱格士菌属(Eggerthella)丰度百分比小于健康对照组,拟杆菌属(Bacteroides)、粪杆菌属(Faecalibacterium)、Subdoligranulum、另枝菌属(Alistipes)、瘤胃球菌属-2(Ruminococcus-2)、假丁酸弧菌属(Pseudobutyrivibrio)、毛螺菌科NK4A136属(Lachnospiraceae_NK4A136_group)、副拟杆菌(Parabacteroides)丰度百分比大于健康对照组。结论非器质性厌食症儿童肠道细菌与健康儿童有差异,Bacteroides含量的增加和Bifidobacterium的减少参与了厌食症的发病。 Objective To study the diversity of the intestinal flora in children with anorexia so as to provide a certain theoretical basis for clinical rational selection of microecological agents.Methods Twenty-four children with anorexia in the children healthcare clinic and pediatric clinic of this hospital from May to October 2018 were set as the anorexia group and 25 children undergoing the healthy physical examination were set as the control group.The stool samples were collected for conducting the bioinformatics analysis after PCR and high-throughput sequencing.Results In the phylum classification level,there were 14 specieses in the stool samples of the anorexia group and 11 specieses in the stool samples of the healthy children group;in the genus level,there were 193 specieses in the stool samples of the anorexia group and 180 specieses in the stool samples of the healthy children group;in the species level,there were 364 specieses in the stool samples of the anorexia group and 326 specieses in the stool samples of the healthy children group.The dominant bacteria in the phylum level among the detected bacteria in the stool samples of the two groups were the same,which were Firmicutes,Bacteroides,Actinomyces,Proteobacteria and Verrucomicrobia.There were 17 dominant bacteria specieses in the genus level.There was no statistical difference in the composition of Firmicutes,Proteobacteria and Verrucomicrobia between the two groups.The content of Bacteroidetes in stool samples of the anorexia group was greater than that in the healthy control group,and the content of Actinomycete in the anorexia group was smaller than that in the healthy control group.There were statistically significant differences in 13 dominant bacterial genera at the genus classification level among the detected bacteria in the stool samples between the two groups(P<0.05).The abundance percentage of Bifidobacterium,Blautia,Streptococcus,Erysipelatoclostridium and Eggerthella in the anorexia group was smaller than that in the healthy control group.The abundance percentage of Bacteroides,Faecalibacterium,Subdoligranulum,Alistipes,Ruminococcus-2,Pseudobutyrivibrio,Parabacteroides and Lachnospiraceae_NK4A136_group in the anorexia group was greater than that in the healthy control group.Conclusion The intestinal bacteria of children with non-organic anorexia are different from the healthy children.The increases in Bacteroides content and decreases in Bifidobacterium participate in the pathogenesis of anorexia.
