摘要
当前肿瘤疾病依然是威胁人们健康、生活质量的一个重要因素,探寻有效的干预手段来缓解肿瘤疾病发展甚至是治愈肿瘤是临床的重要课题,准确的诊断手段、对病情发展与干预程度的评估是控制病情的前提,随着对基因编辑技术了解逐渐深入,近年来尝试将其应用于治疗肿瘤并取得了一定的成效,其中规律成簇间隔短回文重复/CRISPR相关蛋白核酸酶9(Regular cluster interval short palindrome repeats/CRISPR-related protein nuclease 9,CRISPR/Cas9)是相对新型的基因编辑手段,通过该技术不但能够准确完成RNA导向,同时还可实现对于DNA的识别、编辑,这种手段让基因编辑技术在高效、安全等方面得到厚实基础,随着该技术的深入已经实现在碱基水平基础上完成定点修饰基因的操作,为该基因编辑技术治疗肿瘤提供可靠的科学依据。CRISPR/Cas9基因编辑技术近年来被应用于治疗肿瘤,该文就对该技术的分子机制以及对于肿瘤疾病的治疗研究相关进展情况作出综述,为以后临床应用该技术治疗肿瘤疾病提供可参考数据。
Current tumor diseases are still an important factor threatening people’s health and quality of life.Finding effective interventions to alleviate the development of tumor diseases and even cure tumors is an important clinical topic.Accurate diagnosis methods,disease development and intervention levels The evaluation of the disease is the prerequisite for the control of the disease.With the gradual deepening of the understanding of gene editing technology,in recent years,it has tried to apply it to the treatment of tumors and achieved results.Among them,the regular clustered interval short palindrome repeats/CRISPR-related protein nucleic acid Enzyme 9(Regular cluster interval short palindrome repeats/CRISPR-related protein nuclease 9,CRISPR/Cas9)is a relatively new gene editing method.This technology can not only accurately complete RNA guidance,but also realize DNA recognition and editing.This method allows gene editing technology to gain a solid foundation in terms of high efficiency and safety.With the deepening of this technology,it has realized the operation of site-specific modification of genes on the base level,providing reliable science for the gene editing technology to treat tumors in accordance with that CRISPR/Cas9 gene editing technology has been applied to the treatment of tumors in recent years.The paper summarized the molecular mechanism of this technology and related progress in the treatment of tumor diseases,and provided reference data for the future clinical application of this technology in the treatment of tumor diseases.
