基于网络药理学的知母黄柏药对防治糖尿病的作用机制研究 被引量:3

Study on the Mechanism of Rhizoma Anemarrhenae on Prevention and treatment of Diabetes based on Network Pharmacology

摘要 目的探讨知母黄柏药对治疗糖尿病的作用机制。方法通过使用中药系统药理学分析平台(TCMSP)筛选知母和黄柏的活性化学成分,通过TTD和OMIM搜索糖尿病靶点。采用Cytoscape3.7.1软件构建知母黄柏药对-活性化学成分-靶点网络图,将有交集靶标上传到STRING数据库,构建蛋白互作网络图(PPI),并通过DAVID数据库对靶点进行KEGG PATHWAY通路富集分析和Gene Ontology生物学过程富集分析。结果知母黄柏药对-活性化学成分-靶点网络图得到45个活性化学成分作用于89个靶点,主要作用靶点为PTGS2、TNF、PI3K、TLR4,而靶点主要关联糖尿病胰岛素抵抗、胰岛素信号通路、胰高血糖素信号通路、炎症通路和Ⅰ、Ⅱ型糖尿病通路,这些信号通路揭示了知母黄柏药对治疗糖尿病的潜在作用机制。结论从多成分、多靶点、多途径揭示了知母黄柏药对治疗糖尿病的潜在作用机制,为进一步深入研究知母黄柏药对治疗糖尿病作用机制提供了重要的科学和理论依据。 Objective To explore the mechanism of Rhizoma Anemarrhenae in the treatment of diabetes mellitus.Methods The active chemical constituents of Rhizoma Anemarrhenae and Cortex Phellodendri were screened by using traditional Chinese medicine systematic pharmacological analysis platform(TCMSP),and the targets of diabetes were searched by TTD and OMIM.The drug pair-active chemical composition-target network map of Rhizoma Anemarrhenae was constructed by Cytoscape3.7.1 software,the intersecting targets were uploaded to STRING database,and the protein interaction network map(PPI),was constructed.The KEGG PATHWAY pathway enrichment analysis and Gene Ontology biological process enrichment analysis were carried out by DAVID database.Results 45 active chemical constituents acting on 89 targets were obtained from the drug pair-active chemical constituents-target network map of Rhizoma Anemarrhenae,the main targets were PTGS2,TNF,PI3 K and TLR4,.The targets are mainly related to diabetic insulin resistance,insulin signal pathway,glucagon signal pathway,inflammatory pathway and typeⅠandⅡdiabetic pathway,which reveal the potential mechanism of Rhizoma Anemarrhenae in the treatment of diabetes.Conclusion The potential mechanism of Rhizoma Anemarrhenae in the treatment of diabetes is revealed from multi-component,multi-target and multi-way,which provides an important scientific and theoretical basis for further study on the mechanism of Rhizoma Anemarrhenae in the treatment of diabetes.
