摘要
目的:筛选慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)患者外周血单个核细胞中异常表达的lncRNA。方法:收集本院3例COPD病人与3例正常人对照外周血,分离单个核细胞,提取RNA后进行转录组测序,根据P<0.05且|log2FC|>1计算两组样本的表达差异,绘制差异lncRNA热图与火山图。通过Lncpro数据库可能预测差异lncRNA调控的mRNA,并进行GO功能与KEGG信号通路聚类。结果:COPD组和对照组两组间满足P<0.05且|log2FC|>1差异条件的lncRNA共计67个,其中上调33个,下调34个。靶基因主要富集的GO功能:多细胞生物过程的调控功能、发展过程的调控功能、结构体形态发生功能、系统发展功能与发展过程功能。靶基因主要富集的KEGG信号通路:多物种凋亡通路、TGF-β信号通路、补体和凝血级联通路、结肠直肠癌通路与细胞凋亡通路。结论:本研究结果提供了COPD外周血单个核细胞中lncRNA表达变化的一般情况与可能调控功能与通路,可能有助于阐明COPD的潜在机制。
Objective To screen abnormally expressed lncRNAs in peripheral blood mononuclear cells from patients with COPD.Methods The peripheral blood of 3 COPD patients and 3 normal controls was collected from our hospital.Mononuclear cells were isolated,and RNA was extracted for transcriptome sequencing.The expression difference between the two groups of samples was calculated based on P<0.05 and|log2FC|>1.Heat map and volcano map were drawn for difference in lncRNA expression.The Lncpro database was used to predict mRNAs regulated by different lncRNAs,and GO function and KEGG signaling clustering were performed.Results There were 67 lncRNAs between the COPD group and the control group that met the difference of P<0.05 and|log2FC|>1,33 of which were up-regulated and 34 were down-regulated.The target genes were mainly enriched in GO functions regulatory functions of multicellular biological processes and development processes,structural morphogenesis functions,system development functions,and development process functions.Moreover,target genes were mainly enriched in KEGG signaling pathways multi-species apoptotic pathway,TGF-βsignaling pathway,complement and coagulation cascade pathway,colorectal cancer pathway and apoptotic pathway.Conclusion Our results provide general information on changes in lncRNA expression in peripheral blood mononuclear cells of COPD patients along with its possible regulatory functions and pathways,which may help clarify the underlying mechanism of COPD.
