高尔基体整合膜蛋白4在人头颈癌中的表达及临床意义

Expression and clinical significance of golgi integral membrane protein 4 in head and neck cancer

目的探究高尔基体整合膜蛋白4(GOLIM4)在人头颈癌(HNC)中的表达及临床意义。方法选取我院收治的72例头颈部鳞癌(HNSCC)患者的鳞癌组织和癌旁组织分别作为HNSCC组和癌旁组,另选行正常组织37例作为对照组,采用免疫组化、RT-PCR、Western blot法检测各组中GOLIM4 m RNA及蛋白表达情况。结果免疫组化结果显示,GOLIM4主要表达在胞浆中,且在对照组中低表达,在HNSCC组呈现高表达;RT-PCR及Western blot结果显示,HNSCC组中GOLIM4 mRNA及蛋白表达量明显高于对照组,差异具有统计学意义;GOLIM4 m RNA及蛋白表达在病理分级T3-T4级、临床分期Ⅲ-Ⅳ期、淋巴结转移患者中分别高于T1-T2级、Ⅰ-Ⅱ期、无淋巴结转移患者,差异具有统计学意义;随访5年,生存患者GOLIM4 mRNA及蛋白表达水平低于死亡患者,差异具有统计学意义。结论GOLIM4在HNSCC中表达上调,且与患者临床病理及肿瘤转移相关。

Objective To investigate the expression and clinical significance of golgi integral membrane protein 4(GOLIM4)in head and neck cancer(HNC).Methods The squamous cell carcinoma tissues and adjacent tissues from 72 patients with head and neck squamous cell carcinoma(HNSCC)admitted to our hospital were used as the HNSCC group and peritumorial group.Thirty-seven pieces of the normal tissues in head and neck obtained by pathological biopsy or surgery were selected as the control group.Immunohistochemistry,RT-PCR,and Western blot methods were used to detect the expression of GOLIM4 m RNA and protein in each group,and to analyze the relationship with the clinicopathology and prognosis of patients.Spss22.0 software was used for statistical analysis of the data.Results Immunohistochemical results showed that GOLIM4 was mainly expressed in the cytoplasm,and it was low in the control group,and high in the HNSCC group.RT-PCR and Western blot results showed that the expression of GOLIM4 mRNA and protein in the HNSCC group was significantly higher than that in the control group(P<0.05).GOLIM4 mRNA and protein expression was higher in patients with pathological grade T3-T4,clinical stageⅢ-Ⅳ,and lymph node metastasis than those with T1-T2,Ⅰ-Ⅱand no lymph node metastasis(P<0.05).After 5 years of follow-up,the GOLIM4 mRNA and protein expression levels of the surviving patients were lower than those of dead patients(P<0.05).Conclusion The expression of GOLIM4 is upregulated in HNSCC,and related to the clinicopathology and tumor metastasis,which has certain clinical value for the evaluation of patients’condition and prognosis prediction.
