摘要
以贵州六盘水红心猕猴桃果酒为研究对象,对红心猕猴桃果酒提取物清除DPPH自由基和ABTS+自由基的能力进行测定,同时,采用MTS法测定果酒提取物对肝癌HepG2细胞增殖的影响,从而对其抗氧化和抑制人肝癌HepG2细胞活性进行评价。结果表明,猕猴桃果酒提取物对DPPH自由基有一定的清除能力,其IC50值为58.32μg/mL,清除ABTS+自由基IC50值为32.56μg/mL,但其抗氧化能力远弱于阳性对照维生素C。猕猴桃果酒提取物200μM处理5 d后,HepG2细胞活力在70%左右,表明猕猴桃果酒提取物对人肝癌HepG2细胞具有一定的抑制作用,具有潜在抗癌活性,通过激活Nrf2信号通路进而调节抗氧化基因的转录是其可能作用机理。
Kiwifruit wine extract was studied to evaluate its scavenging ability of DPPH and ABTS+.Meanwhile,its anti-proliferation activity of HepG2 cells were studied by MTS method.The results showed that kiwifruit wine extract had certain scavenging ability of DPPH,with an IC50 value of 58.32μg/mL,and its IC50 value was 32.56μg/mL for ABTS+.However,its antioxidant activity was much weaker than that of VC.After treated with 200μM kiwifruit wine extract for 5 days,the viability of HepG2 cells was about 70%,which indicated that kiwifruit wine extract had certain inhibitory effect on HepG2 cells and had potential anti-cancer activity.A possible mechanism is that the activation of Nrf2 signaling pathway regulates the transcription of antioxidant genes.
