摘要
目的:探究马鹿角水提物对秀丽隐杆线虫衰老的影响及可能的抗氧化作用机制。方法:将秀丽隐杆线虫随机分为正常对照组和鹿角水提物1 mg/ml组。观察马鹿角水提物处理后野生型线虫(N2)和突变体线虫(eat-2、daf-2、clk-1)氧化应激条件下存活时间以及野生型线虫(N2)的运动能力、生殖能力和寿命,通过qPCR测定相关抗氧化基因的表达水平。结果:与空白对照组比较,马鹿角水提物1 mg/ml组线虫(N2、eat-2、daf-2)氧化应激条件下的存活时间明显延长(P<0.05或P<0.01),野生型线虫(N2)的运动能力和生殖能力明显提高(P<0.05),但线粒体信号通路突变体线虫clk-1氧化应激条件下的存活时间和寿命无显著差异(P>0.01),N2线虫基因gst-4、gst-7、sod-3和hsp16.2表达水平显著增高(P<0.01)。结论:马鹿角水提物具有显著的体内抗氧化生物活性,能够延缓线虫衰老,其作用机理可能为通过激活线粒体信号通路,上调相关抗氧化基因表达水平,增强线虫抗氧化应激能力,延缓线虫衰老。
Objective:To evaluate the effect of the extract from red deer Cervus elaphus antler on the lifespan of Caenorhabditis elegans(C.elegans)and to explore the mechanism of antioxidant activity.Methods:C.elegans were randomly divided into 1 mg/ml red deer Cervus elaphus antler extract group and control group.After treatment with red deer Cervus elaphus antler extract,the survival time of wild-type worms(N2)and Mutant worms(eat-2、daf-2、clk-1)under oxidative stress was observed,also the motor ability,the reproductive ability and the lifespan of wild type worms(N2)were observed,the expression levels of related oxidant genes were determined by qPCR.Results:Compared with control group,the red deer antler(1 mg/mL)prolonged the survival time of(N2、eat 2、daf 2)worms under oxidative stress(P<0.05 or P<0.01),it also prolonged the motor ability and the reproductive ability in wild type worms(N2)(P<0.05),while no significant difference was observed in lifespan and survival time of clk 1 type mutant worms(P>0.01).The expression levels of gst 4、gst 7、sod 3 and hsp16.2 were increased(P<0.01).Conclusion:The red deer antler has the significant anti-oxidant biological activity in vivo,and can delay the aging of worms.Its mechanism may be related to activating the mitochondrial signal pathway,up-regulating the related antioxidant genes and enhancing the anti-oxidative stress ability of worms.
