高效液相色谱波长切换技术对复方野菊感冒颗粒中8种成分的含量测定 被引量:3

摘要 目的建立同时测定复方野菊感冒颗粒中8个指标成分含量的方法,对复方野菊感冒颗粒进行质量控制研究。方法采用高效液相色谱法结合波长切换技术对复方野菊感冒颗粒中新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C、蒙花苷和甘草酸铵进行定量分析,Boston BosCherom ODS色谱柱(4.6mm×250 mm,5μm),以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,流速1.0 mL·min^-1,0~52 min检测波长为326 nm,52 min后波长切换为250 nm,柱温30℃。结果新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C、蒙花苷和甘草酸铵各组分基线分离,线性范围分别为1.1562~11.5620、1.4100~14.1000、0.9936~9.9360、1.7630~17.6300、0.9936~9.9360、0.8378~8.3780、2.7712~27.7120、4.6028~46.0280μg·mL^-1,呈良好线性关系,r≥0.9999;平均加样回收率分别为100.8%、100.1%、101.2%、101.9%、103.4%、102.6%、101.9%、99.6%,RSD均小于3.0%,3批样品8个指标成分平均含量分别为1.7841、1.7689、1.5114、2.2211、0.8482、0.8286、4.0299、4.7166 mg·袋^-1,不同批次复方野菊感冒颗粒含量差别不大,质量稳定。结论建立的含量测定方法简便准确、重复性好、专属性强,可用于复方野菊感冒颗粒的质量评价。 AIM To establish a quantitative analyses method for determining eight main marker components simultaneously in Fufang Yeju Ganmao granules by HPLC to control its quality.METHODS A high performance liquid chromatography(HPLC)with switching wavelength method was developed for simultaneous determination of eight main components(neochlorogenic acids,chlorogenic acid,cryptochlorogenic acid,isochlorogennc acid B,isochlorogennc acid A,isochlorogennc acid C,linarin and ammonium glycyrrhizinate)on a Boston BosCherom ODS column(4.6 mm×250 mm,5μm)with gradient elution of acetonitrile and 0.1%phosphoric acid as mobile phase at a flow rate of 1.0 mL·min^-1.The wave length was detected at 326 nm between 0 to 52 min,detected at 250 nm after 52 min and the column temperature was maintained at 30℃.RESULTS Neochlorogenic acids,chlorogenic acid,cryptochlorogenic acid,isochlorogennc acid B,isochlorogennc acid A,isochlorogennc acid C,linarin and ammonium glycyrrhizinate showed good linearity in ranger of 1.1562-11.5620,1.4100-14.1000,0.9936-9.9360,1.7630-17.6300,0.9936-9.9360,0.8378-8.3780,2.7712-27.7120 and 4.6028-46.0280μg·mL^-1 with r≥0.9999,and the average recoveries were 100.8%,100.1%,101.2%,101.9%,103.4%,102.6%,101.9%,99.6%with RSD less than 3.0%,respectively.The contents of eight main marker components in 3 batches of Fufang Yeju Ganmao granules were 1.7841,1.7689,1.5114,2.2211,0.8482,0.8286,4.0299,4.7166 mg per bag,respectively.CONCLUSION The established content determination method is simple,accurate,reproducible and specific,which can be used for the quality evaluation of Fufang Yeju Ganmao granules.
