miR-30b和miR-30d对胎盘滋养层细胞HTR8/SVneo迁移能力的影响

Effects of miR-30b and miR-30d on the migration ability of placental trophoblast cells HTR8/SVneo

目的分析miR-30b和miR-30d在子痫前期(PE)胎盘组织中的表达及对胎盘滋养层细胞HTR8/SVneo迁移能力的影响。方法RT-PCR检测10例PE产妇(PE组)及10例正常分娩产妇(对照组)胎盘组织中miR-30b、miR-30d的表达;向滋养层细胞系HTR8/SVneo分别转染miR-30b mimic及miR-30d inhibitor以上调miR-30b和下调miR-30d表达,采用Transwell实验检测转染后各组细胞迁移能力。结果与对照组相比,PE组胎盘组织中miR-30b表达量显著升高,miR-30d显著降低(P<0.05)。Transwell结果显示,与空白组相比,miR-30b-mimic组和miR-30d inhibitor组的细胞迁移数目显著减少(P<0.05)。结论PE胎盘组织中存在miR-30b高表达和miR-30d低表达,其可能通过抑制胎盘滋养层细胞迁移能力参与PE发生发展。

Objective To investigate the expression of miR-30b and miR-30d in PE placental tissue and their effects on the migration ability of placental trophoblast cells HTR8/SVneo.Methods 10 PE puerperas(PE group)and 10 normal labor puerperas(control group)were collected and the contents of miR-30b and miR-30d in placenta tissues were detected by RT-PCR.The expressions of miR-30b in trophoblast cell line HTR8/SVneo was up-regulated by transfection of miR-30b mimic and that of miR-30d was down-regulated by transfection of miR-30d inhibitor,then RT-PCR was used to verify the transfection efficiency,Transwell assay was conducted to detect the migration ability of trophoblast cells in each group.Results Compared with the control group,the expression of miR-30b in PE group was significantly higher and miR-30d was lower(P<0.05).Transwell results showed that the cell migration ability in the miR-30b-mimic and miR-30d inhibitor groups decreased compared with the blank group(P<0.05).Conclusion There was high expression of miR-30b and low expression of miR-30d in placental tissue of PE,which might be involved in the occurrence and development of PE by inhibiting the migration of placental trophoblast cells.
