GTSE1基因在乳头状肾细胞癌中的表达及临床意义 被引量:1

摘要 目的通过数据库探索G2/S期应答相关蛋白1(GTSE1)在乳头状肾细胞癌中的表达及意义。方法通过UALCAN数据库明确GTSE1在乳头状肾细胞癌中的表达差异并进行生存分析,进一步明确GTSE1相关基因,随后利用Metascape工具对相关基因进行通路富集,最后通过OncoLnc数据库对生存分析结果进行验证。结果与正常组相比,GTSE1在乳头状肾细胞癌中高表达(P<0.001),并且以组织亚型、癌症临床分期以及淋巴结转移等作进一步分析后发现,GTSE1在亚组中依然显著高表达(P<0.05),且GTSE1高表达患者生存率显著降低(P<0.01)。同时,乳头状肾细胞癌中GTSE1启动子区甲基化水平显著降低(P<0.001)。通路富集结果表明,GTSE1相关基因主要富集在细胞周期相关通路。结论GTSE1在乳头状肾细胞癌中显著高表达,且与患者不良预后显著相关,其可能通过调控细胞周期来发挥促癌作用。 Objective To explore the expression and significance of G 2 and S phase expressed-1(GTSE1)in papillary renal cell carcinoma(PRCC)by using UALCAN database.Methods Expression of GTSE1 in PRCC was compared using UALCAN database.Survival data of GTSE1 in PRCC was analyzed in UALCAN database and verified by OncoLnc database.Pathway enrichment of related genes carried out by Metascape tool.Results GTSE1 was overexpressed in PRCC compared with normal group(P<0.001).Further analysis of histological subtypes,cancer stage and lymph node metastasis revealed that GTSE1 was still significantly overexpressed(P<0.05),and the survival rate of patients with high GTSE1 expression significantly reduced(P<0.01).Meanwhile,the level of GTSE1 promoter methylation in PRCC was significantly decreased(P<0.001).The pathway enrichment analysis showed that genes associated with GTSE1 were mainly enriched in cell cycle related pathway.Conclusions GTSE1 is up-regulated in PRCC and indicates a worse prognosis.Regulating cell cycle may play a role in promoting cancer.
