响应面法优化鲁氏酵母菌与植物乳杆菌复配培养条件

摘要 试验探索了鲁氏酵母菌与植物乳杆菌的复配培养条件,在单因素试验基础上进行响应面中的Box-Behnken试验设计,以pH和菌落总数为评价指标,优化用于发酵肉制品生产发酵剂的制备条件。结果表明,两组试验的预测模型均极显著,培养温度、培养时间和复配比例间两两交互作用对发酵液pH值和菌落总数影响显著。最终以菌落总数为响应值优化复配培养条件为培养温度20.71℃,培养时间22.99h,Z.rouxii和L.plantarum的复配比例为1.32。对优化结果进行了验证,实际结果与预测值拟合度达94.12%。 In this experiment,the cultivation conditions of Zygosaccharomyces rouxii and Lactobacillus plantarum are tested.On the basis of single-factor test,Box-Behnken experiment design of response surface methodology is carried out to optimize the preparation conditions of starter for fermenting meat products and the pH values and the total number of colonies are used as the evaluation indexes.The results show that the prediction models of the two groups of experiments reach a significant level,and the interactions between the culture temperature,culture time and the compound ratio on the pH values of fermentation broth and the total number of colonies are significant.Finally,the total number of colonies is taken as the response value to optimize the compound culture conditions:the cultivation temperature is 20.71℃,the cultivation time is 22.99 h,and the compound ratio of Z.rouxii and L.plantarum is 1.32.The optimized results are verified,and the fitting degree between the actual values and the predicted values reaches 94.12%.
