摘要
为了提高高产纤维素酶菌株Cladosporium sp.AY-42的产酶能力,采用单因素实验和响应面优化实验对固态发酵产酶条件进行优化。结果表明:培养基的最适碳源为油菜秸秆粉∶麸皮为3∶2,最适氮源为(NH4)2SO4及最适氮源浓度为2%。最佳培养条件为料水比1∶1.8,温度29℃,发酵时间4d,优化后的固态发酵培养菌株CMC酶活最终达(8.17±0.05)IU,酶活力提高了57.72%。该研究将为枝孢菌在纤维素酶发酵生产方面提供一定理论基础。
In order to improve the cellulase production capacity of Cladosporium sp.AY-42,single factor experiment and response surface optimization experiment are used to optimize the enzyme production conditions of solid-state fermentation.The results show that the optimum carbon source of the media is the ratio of rape straw powder to bran is 3∶2,the optimum nitrogen source is(NH 4)2SO 4 and the optimum nitrogen source concentration is 2%.The optimum culture conditions are as follows:the ratio of solid to liquid is 1∶1.8,the temperature is 29℃,and the fermentation time is 4 days.After the optimization,the CMC enzyme activity of the culture strain by solid-state fermentation finally reaches(8.17±0.05)IU,and the enzyme activity is increased by 57.72%.This study will provide a theoretical basis for the fermentation and production of cellulase with Cladosporium.
