近红外免疫层析、胶体金及实时荧光PCR法在A群轮状病毒临床样本检测中的应用

Application of near-infrared immunochromatography,colloidal gold and Real-time PCR in the detection of group A rotavirus in clinical samples

目的评估近红外免疫层析法、胶体金法及实时荧光PCR法在A群轮状病毒临床样本检测中的应用效果。方法采用自主研发的免疫层析试纸条和商品化胶体金及实时荧光PCR试剂盒,检测112份腹泻儿童粪便样本中的A群轮状病毒,比较3种方法的检测结果。结果实时荧光PCR法检测轮状病毒阳性率最高,为77.68%;其次为近红外免疫层析法,阳性率为66.96%;胶体金法阳性率最低,为61.61%。与实时荧光PCR法相比,近红外免疫层析法检测灵敏度为83.91%、特异度为92.00%;胶体金法检测灵敏度为73.56%,特异度为80.00%。在25份实时荧光PCR法检测为阴性样本中,近红外免疫层析法检测出2份阳性,两者符合率为85.71%。结论近红外免疫层析法检测A群轮状病毒的灵敏度高于胶体金法,略低于实时荧光PCR法。

Objective To evaluate the application effect of near-infrared immunochromatography,colloidal gold and Real-time PCR in the detection of group A rotavirus in clinical samples.Methods A total of 112 fecal samples of children with diarrhea were detected by using independently developed immunochromatographic trips,commercial colloidal gold and Real-time PCR kit.Then they were compared with the results of the three methods.Results Realtime PCR had the highest positive rate of 77.68%.The near-infrared immunochromatography was with a positive rate of 66.96%.The colloidal gold had the lowest positive rate of 61.61%.Compared with that of Real-time PCR,the sensitivity and specificity of near-infrared immunochromatography was 83.91%and 92.00%,respectively,while the sensitivity and specificity of colloidal gold was only 73.56%and 80.00%,respectively.Among the 25 samples,2 samples were negative by Real-time PCR but were positive by near-infrared immunochromatography.The coincidence rate was 85.71%between the two methods.Conclusion The sensitivity of near-infrared immunochromatography was higher than that of colloidal gold,and lower slightly than that of Real-time PCR on the detection of group A rotavirus.
