摘要
目的考察苯甲酸雌二醇诱导的小鼠胸腺萎缩模型的稳定性。方法80只雌性ICR小鼠分为对照组与模型组,每组40只。模型组小鼠隔天腹腔注射苯甲酸雌二醇注射液共6次建立胸腺萎缩模型,末次造模给药24h后处理10只小鼠取出胸腺,后每隔7天处理10只小鼠,正常组小鼠除无需造模给药外,其余操作与模型组一致,计算胸腺指数,观察胸腺病理学变化。结果造模结束后,小鼠胸腺体积减小,胸腺指数显著降低;造模结束后1周,小鼠胸腺体积有所恢复,但与正常组相比,胸腺指数有极显著降低;造模结束后2周,小鼠胸腺体积继续恢复,但与正常组相比,胸腺指数有显著降低;造模结束后3周两组别之间无明显差异。病理结果表明,造模结束后3周,模型组小鼠胸腺皮质与髓质分界清晰,较对照组无明显差异。结论苯甲酸雌二醇诱导的雌性小鼠胸腺衰退模型是可逆的,造模后小鼠胸腺萎缩于2周后逐渐恢复至正常。
OBJECTIVE The article is aimed to investigate the stability of thymus atrophy model induced by estradiol benzoate in mice.METHODS Eighty female ICR mice were divided into control group and model group,with 40 mice in each group.In the model group,mice were injected intraperitoneally with estradiol benzoate 6 times every other day to establish a thymus atrophy model.Ten mice in each group were treated 24 hours after the last administration of the model and removed the thymus,and then 10 mice in each group were treated every 7 days.Except that the mice in the control group which did not need to be treated,the rest of the operations were the same as those in the model group,and the thymic index and thymic pathological changes were calculated.RESULTS After treated,the thymus volume of the mice decreased,and the thymus index decreased significantly.The thymus volume of the mice recovered a little after 1 week,but compared with the control group,the thymus index decreased significantly.After 2 weeks,the mouse thymus volume recovered continuely,but compared with the control group,the thymus index was significantly reduced;there was no significant difference between the two groups 3 weeks after treated.The pathological results showed that 3 weeks after treated,the demarcation between thymic cortex and medulla in the model group was clear,and there was no significant difference compared with the control group.CONCLUSION The female mouse thymus regression model induced by estradiol benzoate is reversible,and the mouse thymus atrophy returns to normal after 2 weeks.
