白豆蔻挥发油对庆大霉素所致急性肾损伤大鼠中Caspase-3、Bcl-2、Bax及NF-кB p65蛋白表达的影响

Effect of Amomurn kravanh volatile oil on the expression of Caspase-3,Bax,Bcl-2 and NF-кB p65 in kidney tissues of rats with gentamicin-induced acute kidney injury

目的探讨白豆蔻挥发油对庆大霉素所致大鼠急性肾损伤的保护作用及Caspase-3、Bcl-2、Bax及NF-кB p65蛋白表达的影响。方法将60只SD大鼠随机分为空白对照组、模型组、白豆蔻挥发油低、中、高剂量组阳性药组,每组10只。除空白对照组给予等量生理盐水外,其余各组大鼠腹腔注射庆大霉素(140 mg·kg-1·d-1)诱导急性肾损伤模型,连续7 d;第八天开始灌胃给药,每天1次,连续28 d。收集大鼠尿液、血液、肾组织进行生化指标检测以及对肾组织进行苏木精-伊红(HE)染色;Western blot法用于检测肾组织中Caspase-3、Bcl-2、Bax及NF-кB p65蛋白的表达。结果白豆蔻挥发油能降低急性肾损伤大鼠的尿液中尿蛋白含量和血液中Bun、Scr含量,降低肾体指数(P<0.05)以及改善肾小管病变;能降低急性肾损伤大鼠肾组织中MDA、NO含量和NOS活力,增加肾组织中SOD及GSH-Px活性(P<0.05);能抑制NF-кB p65,Caspase-3,Bax蛋白的表达,促进Bcl-2蛋白的表达(P<0.05)。结论白豆蔻挥发油能改善庆大霉素所致大鼠急性肾损伤,其作用机制与抑制凋亡有关。

Objective To investigate the protective effect of Amomurn kravanh volatile oil on acute kidney injury induced by gentamicin and the expression of Caspase-3,Bcl-2,Bax and NF-кB p65 in rats.Methods Sixty SD rats were randomly divided into normal group,model group,and Amomurn kravanh Pierre ex Gagnep.volatile oil low-dose,medium-dose and high-dose groups,positive group,with 10 rats in each group.Except for normal group,other groups were given gentamicin 140 mg·kg-1·d-1 intraperitoneally to induce acute kidney injury model for 7 days.On the eighth day,oral administration was started,once a day for 28 consecutive days.Rat urine,blood,and kidney tissues were collected for biochemical indicators and hematoxylin-eosin(HE)staining of kidney tissues;Western blot for detection of Caspase-3,Bcl-2,Bax and NF-кB p65 protein expression in kidney tissue.Results Amomurn kravanh volatile oil reduced urine protein content,Bun and Scr content in blood,reduced renal body index(P<0.05)and improved renal tubular lesions in rats with acute kidney injury;Could reduce MDA,NO content and NOS activity,increase SOD and GSH-Px activity in renal tissue of rats with acute kidney injury(P<0.05);Could inhibit the expression of NF-кB p65,Caspase-3,Bax protein,and promote the expression of Bcl-2 protein(P<0.05).Conclusion Amomurn kravanh volatile oil has an improvement effect on acute kidney injury.And the mechanism may be related to inhibition of apoptosis.
