摘要
目的:探讨苦参碱对阿霉素引起的H9c2心肌细胞凋亡的影响。方法:培养H9c2心肌细胞,取23代细胞进行试验,分为4组。对照组:只给生理盐水作为干预因素。阿霉素组:接受浓度1.0mg/L阿霉素作为损伤模型。阿霉素+苦参碱组:接受浓度1.0mg/L阿霉素和150mg/L苦参碱作为干预因素。苦参碱组:接受浓度150mg/L苦参碱作为干预因素。以上各组相应干预24h后,应用MTT法检测H9c2心肌细胞生存率,应用Hoechst33258染色试剂盒检测H9c2心肌细胞凋亡水平,应用激光共聚焦JC-1法检测线粒体膜电位,线粒体呼吸链复合物活性检测试剂盒检测线粒体呼吸链复合物I活性。结果:苦参碱预处理显著提高心肌细胞生存率;与阿霉素组比较,苦参碱能显著降低阿霉素引起的H9c2心肌细胞凋亡(P<0.05),苦参碱能显著改善阿霉素引起的线粒体膜电位降低,与阿霉素组比较有显著差异(P<0.05);与阿霉素组比较,苦参碱能显著提高阿霉素引起的线粒体呼吸链复合物Ⅰ活性的降低(P<0.05)。结论:苦参碱能通过改善线粒体膜电位,提高线粒体呼吸链复合物Ⅰ的活性,减轻阿霉素引起的心肌细胞凋亡提高生存率。
Objective:To investigate the effect of matrine on apoptosis of H9c2 myocardial cells induced by adriamycin.Methods:Cultured H9c2 cardiomyocytes were divided into four groups by taking 2~3 generations of cells for experiment.Control group:only normal saline was used as intervention factor.Adriamycin group:concentration of 1.0mg/L adriamycin was used as intervention factor.Adriamycin+matrine group:concentration of 1.0mg/L adriamycin and 150mg/L matrine were used as intervention factors.Matrine group:150 mg/L matrine was used as the intervention factor.After corresponding intervention for 24h,the survival rate of H9c2 cardiomyocytes was detected by MTT assay,the apoptosis level of H9c2 cardiomyocytes was detected by Hoechst33258 staining kit,the mitochondrial membrane potential was detected by laser confocal JC-1 assay,and the activity of mitochondrial respiratory chain complex I was detected by the mitochondrial respiratory chain complex activity detection kit.Results:Matrine pretreatment significantly improved the survival rate of myocardial cells,and compared with adriamycin group,matrine significantly reduced the apoptosis of H9c2 myocardial cells induced by adriamycin(P<0.05).Compared with the adriamycin group,matrine significantly improved the decreased mitochondrial membrane potential caused by adriamycin,and the difference was statistically significant(P<0.05).Compared with the adriamycin group,matrine significantly increased the reduction of adriamycin induced mitochondrial respiratory chain complex I activity,and the difference was statistically significant(P<0.05).Conclusion:matrine can by improving the mitochondrial membrane potential,increase the activity of the mitochondrial respiratory chain complexⅠ,reduce myocardial cell apoptosis caused by adriamycin,improve myocardial cell survival rate.
