摘要
目的探讨RNA解旋酶Ddx5和Ddx17在儿童髓母细胞瘤组织中的表达及意义。方法免疫组织化学染色法检测68例髓母细胞瘤组织中Ddx5和Ddx17的表达,根据染色结果将患儿分为高表达组和低表达组,比较两组临床病理特征及生存预后。生存率比较采用Log-rank检验;多因素分析采用COX比例风险模型。结果髓母细胞瘤组织中Ddx5低表达17例(25%),高表达51例(75%);Ddx17低表达18例(27%),高表达50例(74%)。Ddx5高表达组和Ddx17高表达组M分期为M1~M4的占比均高于其对应的低表达组(P<0.05)。Ddx5和Ddx17表达均与年龄、性别、肿瘤部位、病理分型无关(P>0.05)。多因素分析结果显示,M分期为M1~M4、Ddx5高表达和Ddx17高表达是髓母细胞瘤患儿不良生存预后的独立影响因素(P<0.05)。Ddx5高表达组中位生存时间为16个月,明显低于Ddx5低表达组的22个月(P<0.05)。Ddx17高表达组中位生存时间为17个月,明显低于Ddx17低表达组的24个月(P<0.05)。结论Ddx5和Ddx17高表达可能预示着髓母细胞瘤患儿的不良生存预后。
Objective To explore the expression and significance of RNA helicases Ddx5 and Ddx17 in children′s medulloblastoma.Methods Immunohistochemical staining was used to detect the expression of Ddx5 and Ddx17 in 68 cases of medulloblastoma.According to the staining results,the children were divided into high expression group and low expression group,and the clinical pathological characteristics and survival prognosis of the two groups were compared.The survival rate was compared using Log-rank test;the multi-factor analysis used COX proportional hazard model.Results In medulloblastoma tissues,17 cases(25%)had low Ddx5 expression,51 cases(75%)had high expression;18 cases(27%)had low Ddx17 expression,and 50 cases(74%)had high expression.Ddx5 high expression group and Ddx17 high expression group,the proportion of M stage M1~M4 were higher than their corresponding low expression group(P<0.05).Both Ddx5 and Ddx17 expression had no relationship to age,sex,tumor location,and pathological classification(P>0.05).The results of multivariate analysis showed that M stages were M1 to M4,high expression of Ddx5 and high expression of Ddx17 were independent influencing factors of poor survival prognosis in children with medulloblastoma(P<0.05).The median survival time of the Ddx5 high expression group was 16 months,which was significantly lower than the 22 months of the Ddx5 low expression group(P<0.05).The median survival time of the Ddx17 high expression group was 17 months,which was significantly lower than the 24 months of the Ddx17 low expression group(P<0.05).Conclusions High Ddx5 and Ddx17 expression may predict the poor survival prognosis of children with medulloblastoma.
