摘要
采用选择性培养基培养的方法,自张掖市临泽县平川镇铁尾矿矿区土壤样品中,分离纯化获得一株对金属铁具有较高耐受性的细菌,命名F1,由形态学特征和16S rRNA序列鉴定其为阴沟肠杆菌(Enterobacter cloacae);菌株培养条件研究表明,接种量1%、培养温度30℃、pH值7.0、NaCl浓度0.5%、摇床转速150 r/min为最适培养条件.重金属耐受性实验显示,该菌株可在浓度分别为1000 mg/L Fe^3+、800 mg/L Pb^2+、150 mg/L Cd^2+、900 mg/L Cu^2+、1100 mg/L Zn^2+的环境中生长,也可在上述五种重金属离子同时存在的环境中生长.Enterobacter cloacae F1表现出了良好的重金属耐受性,具备对重金属污染环境进行生物修复的潜能.
In this paper,a bacterium with high resistance to iron was isolated and purified from soil samples of iron tailings mining area in Pingchuan Town,Linze County,Zhangye City by selective culture medium method and was named F1.After morphological identification and 16S rRNA gene sequence determination,the strain was confirmed as Enterobacter cloacae.The optimum culture conditions were the inoculation amount of 1%,culture temperature of 30℃,pH value of 7,NaCl concentration of 0.5%,and shaking speed of 150 r/min.The results of heavy metal tolerance test showed that the strain could grow well in 1000 mg/L Fe^3+、800 mg/L Pb^2+、150 mg/L Cd^2+、900 mg/L Cu^2+、1100 mg/L Zn^2+,respectively,and also could grow well in the allied presence of the above five heavy metal ions.Enterobacter Cloacae F1 demonstrated better tolerance to heavy metals and possessed the potential for bioremediation of heavy metal pollution.
