摘要
试验旨在研究利巴韦林及其主要代谢物1H-1,2,4-三氮唑-3-甲酰胺(TCONH2)在肉鸡组织中的残留消除规律。90只矮脚黄鸡随机分为9组,根据10 mg/kg体重剂量连续7 d口服利巴韦林溶液。停药后采集胸肉、腿肉和肝脏样品,采用改进的UPLC-MS/MS方法进行检测。结果显示:该方法操作简单、耗时短、回收率高,可与内源性干扰物尿苷分开,能对低水平利巴韦林和TCONH2进行定量分析检测;胸肉、腿肉和肝脏中利巴韦林和TCONH2平均加标回收率分别为102.2%~110.2%和91.8%~112.6%,相对标准偏差范围为2.4%~7.9%;当停药0.25 d后,利巴韦林和TCONH2在胸肉、腿肉和肝脏中残留量最高,且在肝脏中消除速率较慢,停药21 d后仍可在肝脏中检出TCONH2残留。研究提示肝脏是监测TCONH2残留的最佳靶组织,结果对于深入了解利巴韦林在肉鸡体内残留代谢规律和提供畜禽产品质量安全监管方法具有十分重要的参考价值。
A method for determination of ribavirin and its metabolite 1 H-1, 2, 4-triazole-3-formamide(TCONH2) in tissues of broilers was established. Ninety yellow dwarf chicken were selected as experimental animals and divided into nine groups. Ribavirin solutions were taken orally for 7 consecutive days at dose of 10 mg/kg according to body weight. The tissue samples of breast muscle, leg muscle and liver were collected after the withdrawal period, and the improved UPLC-MS/MS method was used for determination. The ribavirin and TCONH2 were detected by improved UPLC-MS/MS with positive electrospray ionization-mass spectrometry in the multiple reaction monitoring mode and quantified with13 C5-ribavirin as the internal standard. The method was simple to operate, less time-consuming, high recovery rate, and could distinguish the endogenous interfering uridine and used for quantitative analysis and detection of low levels of ribavirin and TCONH2;Recoveries of ribaverin and TCONH2 in breast muscle, leg muscle and liver were 102.2% to 110.2% and 91.8%% to 112.6% separately, with a relative standard deviation ranged from 2.4% to7.9%. A quarter of day after the withdrawal period, the residual levels in breast muscle, leg muscle and liver reached the highest level, and the elimination rate of ribavirin and TCONH2 in liver was relately slow. TCONH2 residues could still be detected in the liver after 21 d, and liver could be used as the residual target tissue for monitoring the metabolite TCONH2. The results were of great value for further understanding the residue metabolism of ribavirin in broiler chickens and provided the methods for quality and safety supervision of livestock and poultry products.
