雷公藤多甙对骨缺损模型大鼠Foxp3及骨形态发生蛋白7表达的影响 被引量:2

Effects of Tripterygium wilfordii polyglycosides on the expression of Foxp3 and bone morphogenetic protein 7 in bone defect model rats

摘要 目的探讨雷公藤多甙对骨缺损模型大鼠叉头框蛋白P3(Foxp3)及骨形态发生蛋白7(BMP7)表达的影响。方法清洁级雌性SD大鼠36只,随机数字表法分为2组:雷公藤多甙组与对照组,每组各18只。2组均建立骨缺损模型,雷公藤多甙组给予雷公藤多甙20 mg处理缺损部位,对照组于缺损部位放入等体积的小牛血清。术后6周与12周,记录2组大鼠形态变化、最大载荷与最大应变量;测量椎体骨组织学形态参数,包括骨小梁体积(TBV)百分比和骨小梁表面(TRS)百分比;免疫组化分析Foxp3和BMP7表达情况;观察2组大鼠病理组织情况。结果所有大鼠均顺利建模成功,精神状况良好,反应灵敏、迅速,饮食、饮水均正常,毛发整洁。术后6周与12周,雷公藤多甙组大鼠的最大载荷、最大应变量均显著高于对照组,差异有统计学意义(P<0.05)。雷公藤多甙组大鼠TBV%显著高于对照组,TRS%显著低于对照组,差异有统计学意义(P<0.05)。雷公藤多甙组大鼠Foxp3、BMP-7相对表达水平均显著高于对照组,差异有统计学意义(P<0.05)。雷公藤多甙组缺损部位病理病变较对照组轻,炎性细胞浸润明显减少,软骨基质及软骨细胞无退行性变,关节软骨表面光滑。结论雷公藤多甙在骨缺损模型大鼠中的应用安全性佳,能促进Foxp3及BMP-7的表达,可改善骨缺损组织的组织学形态与椎体力学状况,有利于提高修复效果。 Objective To investigate the effect of Tripterygium wilfordii polyglycosides on the expression of P3(Foxp3)and bone morphogenetic protein(BMP)7 in bone defect model rats.Methods 36 cases of clean-grade female SD rats were randomly divided into two groups by number table,18 cases in each of the Tripterygium wilfordii polyglycosides group and the control group.Bone defect models were established in both groups.The Tripterygium wilfordii polyglycosides group were given tripterygium wilfordii polyglycoside 20 mg to treat the defect site,and the control group were placed equal volume of calf serum in the defect.At 6 weeks and 12 weeks after surgery,the morphological changes,maximum load and maximum strain of the two groups of rats were recorded;the histological parameters of the vertebral body were measured,including the percentage of trabecular bone volume(TBV)and the trabecular bone surface(TRS))Percentage;Foxp3 and BMP7 expression were analyzed by immunohistochemistry;the pathological tissue of rats in the 2 groups was observed.Results All rats were successfully modeled successfully,with good mental state,responsive and rapid reaction,normal diet and drinking water,and clean hair.At 6 weeks and 12 weeks after surgery,the maximum load and maximum strain in the Tripterygium wilfordii polyglycosides group were significantly higher than those of the control group,the differences were statistically significant(P<0.05).The percentage of trabecular bone volume in the Tripterygium wilfordii polyglycosides group were significantly higher than that of the control group,trabecular bone the surface percentage were significantly lower than the control group,the differences were statistically significant(P<0.05).The relative expression levels of Foxp3 and BMP-7 in the Tripterygium wilfordii polyglycosides group were significantly higher than the control group,the differences were statistically significant(P<0.05).The pathological lesions in the defect area of the Tripterygium wilfordii polyglycosides group were lighter than those of the control group,the infiltration of inflammatory cells were significantly reduced,the cartilage matrix and chondrocytes were not degenerated,and the surface of the articular cartilage were smooth.Conclusion Tripterygium wilfordii polyglycoside has good safety in the rat model of bone defect,can promote the expression of Foxp3 and BMP-7 and improve the histological morphology of the bone defect tissue and the mechanical status of the vertebral body.It is beneficial to improve the repair effect.
