摘要
基于LC-MS/MS代谢组学方法对水培提取、原位提取、鲜土浸提提取和风干土提取等4种方法提取的槟榔根系分泌物进行定性和定量分析,对比不同提取方法提取的差异物质。结果表明:水培提取、原位提取、鲜土提取和风干土提取等4种方法分别检测到438种、401种、369种和346种代谢物,4种提取方法所获得的主要代谢物为氨基酸、核苷酸、有机酸、黄酮类、脂质,占到所有检测到的代谢物的62.9%~64.2%;进一步对比发现,水培提取方法提取的物质丰富度最高,原位提取的物质最接近自然条件下植物根系分泌物组分;鲜土提取较原位提取,主要损失的代谢物为脂质类、核苷酸类和黄酮类,而风干土提取主要损失的代谢物为有机酸类和氨基酸类。因此,在开展根系分泌物研究时,应根据试验目的选择适宜收集方法。
Betel nut root exudates were extracted by using four methods,hydroponic extraction,in situ extraction,fresh soil extraction and air-dried soil extraction,and were qualitatively and quantitatively determined by using the LC-MS/MS.The components of the extracts were compared.The results showed that the root exudates contained 438,401,369 and 346 metabolites,respectively when detected by using the four methods.The main metabolites derived from the root exudates extracted by the four extraction methods are amino acids,nucleotides,organic acids,flavonoids,and lipids,accounting for 62.9%-64.2%of all the detected metabolites.Further comparison found that the contents of lipids,nucleotides and flavonoidsthe decreased in the samples of the fresh soil extraction compared to those in the in situ extraction,while the contents of organic acids and amino acids decreased in the samples of the air-dried soil extraction compared to those in the fresh soil extraction.The components of the root exudates are most abundant in the samples of the hydroponic extraction,while the components in the samples of the in-situ extraction are more close to those of the plant root exudates under natural conditions.Therefore,an appropriate extraction method for root exudates should be selected according to the experimental purpose.
