摘要
对基于核酸适体酶联核酸适配体法(酶联适体法)、核酸适配体结构转换荧光法(适体转换荧光法)和高效液相色谱法(HPLC)检测黄曲霉毒素B 1进行对比研究。结果表明:当花生加标量为5、20、60μg/kg时,酶联适体法测得4.27、18.24、56.66μg/kg,适体转换荧光法测得4.95、21.05、65.00μg/kg,高效液相色谱法测得4.62、16.90、48.00μg/kg;当玉米加标量为5、20、60μg/kg时,酶联适体法测得4.05、17.34、50.79μg/kg,适体转换荧光法测得4.05、17.11、50.77μg/kg,高效液相色谱法测得5.10、16.33、44.98μg/kg。统计学分析结果显示,两种基于核酸适体的方法与高效液相色谱法两两之间无显著性差异(P>0.05),且酶联适体法的检测结果具有可视化,核酸适体结构转换荧光法更为直观快捷,为探索黄曲霉毒素B 1的快速检测方法奠定了基础。
This study compared the effects of aflatoxin B 1(AFB 1)detection methods:nucleic acid aptamerbased enzyme-linked aptamer(enzyme-linked aptamer),nucleic acid aptamer structure conversion fluorescence(aptamer conversion fluorescence),and high performance liquid chromatography(HPLC).The results showed that when the additions of AFB 1 in peanuts were 5,20,and 60μg/kg,the detection results by enzyme-linked aptamer were 4.27,18.24,and 56.66μg/kg,respectively,the detection results by conversion fluorescence were 4.95,21.05,and 65.00μg/kg,respectively,and the detection results by HPLC were 4.62,16.90,and 48.00μg/kg,respectively.When the additions of AFB 1 in corns were 5,20,and 60μg/kg,the detections results by enzyme-linked aptamer were 4.05,17.34,and 50.79μg/kg,respectively,the detections by conversion fluorescence were 4.05,17.11,and 50.77μg/kg,respectively,and the detection results by HPLC were 5.10,16.33,and 44.98μg/kg,respectively.Statistical analysis showed that either two of the three methods mentioned above had no significant difference(P>0.05).Among them,enzyme-linked aptamer method was better visualized,and aptamer structure transformation fluorescence method was more intuitive and quick,which provided the base for the fast AFB 1 detection method.
