肉桂、知母对寒热体质大鼠下丘脑TRP通道的调整作用 被引量:4

Adjustment Effect of Rougui(cinnamon) and Zhimu(Rhizoma anemarrhenae) on TRP Channel of Hypothalamus in Rats with Cold and Heat Constitutions

摘要 目的依据“寒者热之,热者寒之”理论,研究利用热性中药肉桂、寒性中药知母分别对寒热体质大鼠进行体质调整,通过观察体质调整前后,常体、寒体和热体体质大鼠下丘脑TRP通道的表达、掌温和热痛阈的变化,探讨中药对寒热体质调整作用及作用机制,为临床偏颇体质的调整、实现“未病先防”提供科学依据。方法以掌温结合自主活动,从自然群体大鼠筛选出常体、热体及寒体大鼠,再将各体质大鼠分成体质未调整组与体质调整组。体质调整组寒体、热体大鼠分别给予肉桂、知母,常体大鼠给予相应体积的饮用水,连续灌胃12周。测量体质未调整组和体质调整组大鼠掌温、热刺痛仪测定大鼠热痛阈、RT-PCR和Western Blot技术检测大鼠下丘脑TRPV1和TRPV4通道表达。结果体质未调整组寒体大鼠掌温显著低于热体大鼠和常体大鼠,而热痛阈显著高于其余两种体质大鼠,掌温和热痛阈呈负相关;寒体体质大鼠下丘脑TRPV1和TRPV4通道表达低于热体体质大鼠。体质调整组大鼠,经过相应药物对体质进行调整后,常体、寒体和热体体质大鼠掌温和热痛阈之间差异性减小,差异无统计学意义;寒体组大鼠下丘脑TRPV1、TRPV4通道表达出现上升趋势,而热体组大鼠呈下降趋势,与常体组差异无统计学意义。结论肉桂、知母对寒热体质大鼠体质调整作用,可能与调节下丘脑TRP通道的表达有关。 Objective According to the theory of "treating cold syndrome with heat drugs andtreating heat syndrome with cold drugs",this study utilized cold drug Zhimu(Rhizoma anemarrhenae)and heat drug Rougui(cinnamon) to identify the rats with cold and heat constitutions,to observe the expression of TRP channel in the hypothalamus of rats and the changes of palmar temperature and thermal pain threshold and to explore traditional Chinese medicine regulating cold and heat constitutions and its mechanism to provide a scientific basis for the adjustment of clinical biased constitutions and the realization of "preventing disease before onset". Methods The rats in the natural population were screened for rats with normal, heat and cold constitutions by combining palm and temperature with autonomic activities. The rats of each constitution were divided into the unadjusted group and theadjustment group. The adjustment group of rats with cold and heat constitutions were given Rougui(cinnamon) and Zhimu(Rhizoma anemarrhenae)respectively, and the normal ratswere given corresponding volume of drinking water for 12 weeks. Rats in the unadjusted group and theadjustment group were measured the thermal pain threshold. RT-PCR and Western Blot were used to detect the expressions of TRPV1 and TRPV4 channels in rat hypothalamus. Results In the unadjusted group, the palm temperature of rats with cold constitution was significantly lower than that of the rats with heat and normalconstitutions and the thermal pain threshold was significantly higher than that of the rats with other two constitutions. The palm temperature and thermal pain threshold were negatively correlated. The expressions of TRPV1 and TRPV4 channels in rat hypothalamus were lower than that in rats with heat constitution. In theadjustment group, the difference between the palm warm and thermal pain thresholdswas reduced, and the difference was not significant. The coldconstitution group’s expressions of TRPV1 and TRPV4 channels showed an upward trend, while theheat constitutiongroup showed a downward trend, which was not significantly different from the normalconstitutiongroup. Conclusion The effects of Rougui(cinnamon) and Zhimu(Rhizoma anemarrhenae)onrats with cold and heat constitutions may be related to the regulation of the expressions of TRP channels in the hypothalamus.
