摘要 目的研究蒙古扁桃药材石油醚提取物对肾纤维化大鼠的保护作用。方法采用单侧输尿管结扎方法建立肾纤维化模型。将实验动物分为UUO模型组、假手术组、阳性药盐酸贝那普利组(1.5 mg·kg^-1)、蒙古扁桃石油醚提取物高剂量组(1.75 g·kg^-1)、中剂量组(1.25 g·kg^-1)和低剂量组(0.75 g·kg^-1)。造模同时灌胃给药21 d后,统计大鼠尿量,计算大鼠脏器指数,测定大鼠血清中尿素氮(BUN)、肌酐(Scr)、白蛋白(ALB)、超氧化物歧化酶(SOD)、丙二醛(MDA)和肾脏组织中SOD、MDA、羟脯胺酸(HYP),观察肾脏组织学病理变化。结果与模型组比较,蒙古扁桃药材石油醚提取物可降低脏器指数,降低血清尿素氮、肌酐、白蛋白水平、降低血清MDA及组织MDA、HYP含量,可提高血清SOD及组织SOD活力,差异均有统计学意义(P<0.05)。肾组织切片HE、Masson染色结果也表明,蒙古扁桃石油醚提取物减缓了肾纤维化程度。结论实验结果表明蒙古扁桃药材石油醚提取物对单侧输尿管结扎法所致的大鼠肾纤维化具有良好的保护作用。 Objective To study the protective effect of Amygdalus mongolica oil extract on renal fibrosis in rats.Methods The renal fibrosis model was established by unilateral ureteral ligation.The experimental animals were divided into UUO model group,sham operation group,positive benazepril hydrochloride group(1.5 mg·kg^-1),Amygdalus mongolica oil ether extract high dose group(1.75 g·kg^-1),medium dose group(1.25 g·kg^-1)and low dose group(0.75 g·kg^-1).The urine volume of the rats was calculated,and the organ indexes were calculated.The serum levels of urea nitrogen(BUN),creatinine(Scr),albumin(ALB),superoxide dismutase(SOD),malondialdehyde(MDA)and SOD,MDA and HYP were measured.Results The results showed that compared with the model group,Amygdalus mongolica oil ether extract could reduce the organ index,decrease the levels of serum urea nitrogen,creatinine and albumin,decrease the contents of serum MDA and tissue MDA and HYP,and increase serum SOD and tissue SOD activity,with significant differences(P<0.05).The results of HE and Masson staining of kidney tissue sections also showed that Amygdalus mongolica oil ether extract reduced the degree of renal fibrosis.Conclusion The results show that Amygdalus mongolica oil ether extract has a good protective effect on rat renal fibrosis induced by unilateral ureteral ligation.
