大孔树脂法富集黑骨藤总黄酮工艺研究 被引量:5

Study on Extraction and Purification of Total Flavonoids from Periploca Forrestii Schltr by Macroporous Resin

摘要 目的优选大孔树脂分离纯化黑骨藤总黄酮的最佳工艺参数。方法采用紫外分光光度法测定黑骨藤总黄酮含量,利用静态及动态吸附解吸试验考察5种不同型号的大孔树脂对黑骨藤总黄酮的吸附及解吸能力,选出适宜的大孔树脂,最后确定其最佳分离纯化条件。结果D101型大孔树脂对黑骨藤总黄酮有较好的吸附率和解析附能力,最佳分离纯化工艺条件为:黑骨藤上样液浓度33.34 mg/mL,上样量0.58 BV,静置时间为24 h,水洗体积为2.94 BV,洗脱流速2 BV/h,洗脱溶剂用70%乙醇,洗脱溶剂用量5.88 BV。结论D101型大孔树脂在所得工艺条件下可以对黑骨藤中总黄酮进行较好的分离纯化,可用于黑骨藤总黄酮的分离纯化。 Objective To optimize the optimal process parameters for separation and purification of total flavonoids from Periploca forrestii Schltr with macroporous resin.Methods UV spectrophotometry was used to determine the contents of total flavonoids in Periploca forrestii Schltr Static and dynamic adsorption and desorption tests were used to investigate the adsorption and desorption capacity of five different types of macroporous resins to total flavonoids in Periploca forrestii Schltr,and the appropriate macroporous resins were selected.Finally,the optimal separation and purification conditions were determined.Results D101 macroporous resin had a good adsorption rate and desorption capacity for total flavonoids from Periploca forrestii Schltr.The optimal separation and purification conditions were as follows:concentration of Periploca forrestii Schltr loading solution 33.34 mg/mL,loading amount 0.58 BV,standing time 24 hours,washing volume 2.94 BV,elution flow rate 2 BV/h,elution solvent 70% ethanol,elution solvent dosage 5.88 BV.Conclusion D101 macroporous resin can be used for the separation and purification of total flavonoids from Periploca forrestii Schltr under the obtained conditions,and can be used for the separation and purification of total flavonoids from Periploca forrestii Schltr.
