摘要
中医治疗疾病以中药方剂为载体,方剂由多味中药构成,针对特定疾病的中药处方集开展中药网络分析有助于探索组方规律和作用机制。收集国医大师吕仁和教授372首治疗肾病综合征(Nephrotic Syndrome,NS)处方,采用Apriori关联规则方法分析常用药物、药对和药组,根据药物处方共现关系和频度构建加权药物网络,引入改进互信息方法提取核心药物,开展核心中药成分靶点预测,结合NS相关基因构建中药-成分-靶点-疾病基因网络,探索中药治疗NS的有效成分和作用机制。实验结果表明吕仁和教授治疗NS常用药物为猪苓、益智仁、羌活等,常用药对为羌活-猪苓、益智仁-猪苓等,基因本体富集分析核心药物成分靶点的富集GO项与NS基因的富集GO项高度吻合,表明吕仁和教授治疗NS处方中核心药物的有效性和合理性。
In traditional Chinese medicine(TCM),diseases are treated by prescriptions composed of several Chinese herbs.The herb network analysis for the prescription set of a specified disease is helpful to explore its composition principle and mechanism.In this paper,we collected 372 prescriptions for nephrotic syndrome(NS)treatment from a TCM master,Professor LYU Renhe,analyzed his commonly-used Chinese herbs,herb pairs and groups by Apriori association rule,and built a weighted herb network based on the co-occurrence relation and frequency of herbs in the given prescriptions.After that,we extracted the core herbs by using an improved mutual information method,predicted the targets of core herbs’components,and explored the effective components and their mechanism for NS treatment by building the herb-component-target-gene network.Experimental results showed in the prescriptions for NS treatment of Professor LYU,the commonly-used herbs included Zhuling(Polyporus umbellatus),Yizhiren(Alpinia oxyphylla),Qianghuo(Notopterygium incisum)and so on.And the commonly-used herb pairs included Qianghuo(Notopterygium incisum)-Zhuling(Polyporus umbellatus),Yizhiren(Alpinia oxyphylla)-Zhuling(Polyporus umbellatus),etc.In addition,it is found that enrichment GO items of targets of core herbs’components are significantly overlapped with enrichment GO items of NS genes,indicating the validity and rationality of Professor LYU’s core herbs for NS treatment.
