摘要
目的研究miR-590-5p在乳腺癌中的表达情况,探讨miR-590-5p对乳腺癌细胞SK-BR-3生物学行为的影响,明确miR-590-5p在乳腺癌细胞中的作用。方法选取2015年1月至2019年1月重庆市北碚区中医院诊治的30例乳腺癌患者的癌组织和其对应的癌旁正常组织作为研究材料。应用荧光定量聚合酶链反应(Real-time PCR)技术研究miR-590-5p在30例乳腺癌癌组织及其癌旁正常组织中的表达情况,体外培养SK-BR-3细胞进行转染。将其分两组,分别为实验组和阴性对照组。利用CCK8实验观察乳腺癌细胞SK-BR-3在0h、24h、48h、72h生长增殖情况;细胞划痕愈合试验观察乳腺癌细胞SK-BR-3在24h、48h的迁移情况;利用Transwell试验观察乳腺癌癌细胞SK-BR-3侵袭能力。Western blot检测MMP-2和MMP-9蛋白的表达情况。结果miR-590-5p在乳腺癌组织中表达低于癌旁正常组织。过表达miR-590-5p后,实验组与阴性对照组相比,细胞生长增殖受抑;划痕试验显示过表达miR-590-5p后实验组迁移距离远远小于阴性对照组;Transwell结果表明实验组细胞穿膜数量显著小于阴性对照组。Western blot结果显示实验组MMP-2和MMP-9蛋白的表达量均低于阴性对照组。结论过表达miR-590-5p抑制SK-BR-3细胞生长增殖,同时也抑制SK-BR-3细胞的侵袭迁移能力,并且很可能通过下调MMP-2和MMP-9蛋白的表达来抑制乳腺癌细胞的侵袭迁移。
Objective To investigate the expression of miR-590-5p in breast cancer and explore the effect of miR-590-5p on the biological behavior of breast cancer cell SK-BR-3,and to clarify the role of miR-590-5p in breast cancer cells.Methods The breast cancer tissues and non-breast cancer tissues of 30 patients were collected from January 2015 to January 2019 in Hospital of Beibei District Traditional Chinese Medicine,Chongqing.Realtime polymerase chain reaction(PCR)was used to study the expression of miR-590-5p in 30 breast cancer tissues and adjacent normal tissues.SK-BR-3 cells were divided into test group and control group respectively.The growth and proliferation of breast cancer cells at 0 h,24 h,48 h and 72 h were observed by CCK8.Cell scratch healing and Transwell experiment were used to observe the invasion ability of breast cancer cells.The expression of MMP-2 and MMP-9 were detected by Western blot.Results The expression of miR-590-5p in breast cancer tissues was lower than that in normal adjacent tissues.After overexpression of miR-590-5p,cell proliferation was inhibited in test group compared with the control group.The results showed that the migration distance of test group was significantly lower than that of the control group after the expression of miR-590-5p.Transwell results indicated that the number of cells in the test group was significantly smaller than that of the control group.Western blot showed that the expression levels of MMP-2 and MMP-9 in test group were lower than those in the control group.Conclusions Overexpression of miR-590-5p not only inhibits the cells proliferation of SK-BR-3 but also the migration and invasion in SK-BR-3 cells,which is likely to inhibit breast cancer cell migration through decreasing the expression of MMP-2 and MMP-9.
