摘要
目的探讨维生素D对体外成熟小鼠卵母细胞发育潜能的影响。方法采集小鼠生发泡(GV)期卵母细胞进行体外成熟培养,根据培养液的不同分为对照组(采用IVM培养基,取卵数188个)和实验组(在对照组基础上添加固体维生素D,取卵数123个)。所有细胞体外进行培养,记录并分析不同条件下处理下两组卵母细胞成熟率、孤雌激活率、囊胚形成率以及表观遗传相关基因表达水平情况。结果两组MII卵母细胞率、孤雌激活率比较,差异无统计学意义(P>0.05)。实验组囊胚形成率显著高于对照组,差异具有统计学意义(P<0.05)。与对照组相比,实验组所选13个基因(Mat2a、Dnmt1、Dnmt3a、Dnmt3b、Dnmt3L、Mbd1、MeCP2、Mbd3、Ezh2、Suz12、Phc2、Rnf2和YY1)表达明显高于对照组,差异具有统计学意义(P<0.05)。结论维生素D可促进卵母细胞在体外成熟,可以提高卵母细胞的发育潜能。
Objective To investigate the effect of vitamin D on the developmental potential of oocytes in vitro mature mice.Methods Mouse oocytes at GV stage were collected for in vitro mature culture,and were divided into control group(IVM medium,number of eggs=188)and experimental group(vitamin D was added to the control group,number of eggs=123)according to different culture medium.All cells were cultured in vitro,and the maturation rate,parthenogenetic activation rate,blastocyst formation rate and epigenetic gene expression levels of oocytes in the two groups were recorded and analyzed under different conditions.Results There was no significant difference in MII oocyte rate and parthenogenetic activation rate between the two groups(P>0.05).Blastocyst formation rate in the experimental group was significantly higher than that in the control group,and the difference was statistically significant(P<0.05).Compared with the control group,the expressions of 13 selected genes(Mat2 a,Dnmt1,Dnmt3 a,Dnmt3 b,Dnmt3 L,Mbd1,MeCP2,Mbd3,Ezh2,Suz12,Phc2,Rnf2 and YY1)in the experimental group were significantly higher than those in the control group,with statistically significant differences(P<0.05).Conclusions Vitamin D can promote oocyte maturation in vitro and improve the development potential of oocytes.
