摘要 肌动蛋白纤维相关蛋白1-反义RNA 1(AFAP1-AS1)为一个新发现的促癌长链非编码RNA(lnc RNA)。AFAP1-AS1定位于人类4号染色体4p16.1区域,被证实在食管癌、胰腺癌、鼻咽癌、肺癌、卵巢癌、结直肠癌、胆道癌、胃癌、肝细胞癌、乳腺癌、舌鳞状细胞癌、视网膜母细胞瘤、胶质瘤和骨肉瘤等多种肿瘤组织中表达上调。AFAP1-AS1过表达与肿瘤大小、淋巴转移、远处转移、TNM分期及不良预后等恶性肿瘤临床病理特征密切相关。此外,AFAP1-AS1可通过调控AFAP1和EMT相关基因、Rho/Rac代谢通路、PTEN/p-AKT通路等促进肿瘤细胞增殖、迁移和侵袭,影响细胞周期进程及抑制细胞凋亡等。 Actin fiber-associated protein 1-antisense RNA 1(AFAP1-AS1)is a newly discovered cancer-promoting long non-coding RNA(lnc RNA).AFAP1-AS1 is mapped to the 4 p16.1 region of chromosome 4 and found to be upregulated in many malignancies,including esophageal cancer,pancreatic carcinoma,nasopharyngeal carcinoma,lung cancer,ovarian cancer,colorectal cancer,biliary tract cancer,gastric cancer and hepatocellular carcinoma,breast cancer,tongue squamous cell carcinoma,retinoblastoma,glioma and osteosarcoma.Overexpression of AFAP1-AS1 was shown to be associated with several clinicopathological features such as tumor size,lymphatic metastasis,distant metastasis,TNM stage and poor prognosis.Additionally,AFAP1-AS1 may promote cell proliferation,migration and invasion,regulate cell cycle progress and inhibit apoptosis through modulating the expression of AFAP1,EMT related genes,metabolic pathway of Rho/Rac and PTEN/p-AKT pathway.
