蒙药蓝刺头对绝经后骨质疏松症模型大鼠骨密度、骨形态发生蛋白-2及Lee’s指数的影响 被引量:1

Effects of Mongolian medicine Echinops latifolius Tausch on bone mineral density,bone morphogenetic protein-2 and Lee’s index in postmenopausal osteoporosis model rats

摘要 目的观察蒙药蓝刺头对绝经后骨质疏松症(PMOP)模型大鼠骨密度(BMD)、骨形态发生蛋白-2(BMP-2)及Lee’s指数的影响。方法将56只大鼠按照随机数字表法分为假手术组,模型组,己烯雌酚组,强骨胶囊组,蓝刺头高、中、低剂量组7组,每组8只。假手术组不切除卵巢,仅切除卵巢周围少量脂肪;其余各组均切除卵巢建立PMOP大鼠模型,90 d后各组分别给予相应药物灌胃,每日1次,连续灌胃90 d。比较各组模型期和治疗期的Lee’s指数,末次给药后的右侧股骨干骺端BMD及骨小梁体积分数(BV/TV);采用免疫组化染色,测定BMP-2光密度值,并进行比较。结果与假手术组比较,模型组BMD、BV/TV及BMP-2光密度值均明显降低,差异有统计学意义(P<0.01);与模型组比较,己烯雌酚组、强骨胶囊组及蓝刺头各剂量组BMD、BV/TV均明显升高,己烯雌酚组、强骨胶囊组及蓝刺头中剂量组BMP-2光密度值均明显升高(P<0.05或P<0.01)。与假手术组比较,模型组模型期及治疗期体质量、Lee’s指数均明显升高,差异有统计学意义(P<0.05或P<0.01);与模型组比较,己烯雌酚组、强骨胶囊组及蓝刺头各剂量组治疗期Lee’s指数均明显降低,差异有统计学意义(P<0.05)。结论蒙药蓝刺头能明显升高PMOP模型大鼠BMD、BV/TV及BMP-2表达,降低Lee’s指数,从而有效抗PMOP。 Objective To observe the effects of Mongolian medicine Echinops latifolius Tausch on bone mineral density(BMD),bone morphogenetic protein-2(BMP-2) and Lee ’s index in postmenopausal osteoporosis(PMOP) model rats. Methods Fifty-six rats were divided into 7 groups according to random number table method,including sham operation group,model group,diethylstilbestrol group,Qianggu Jiaonang(强骨胶囊)group,Echinops latifolius Tausch high,medium and low dose group,with 8 rats in each. In the sham operation group,only a small amount of fat around the ovaries was removed;the other groups were ovariectomized to establish PMOP model rats. After 90 days,each group was given corresponding drugs by gavage once a day for 90 days. The Lee’s index during the model period and the treatment period among each group,BMD of right femoral metaphysis and bone trabecular volum fraction(BV/TV) after the last administration were compared. Immunohistochemical staining was used to determine the optical density of BMP-2 and was compared. Results Compared with the sham operation group,BMD,BV/TV and BMP-2 optical density in the model group significantly decreased,and the difference was statistically significant(P<0.01).Compared with the model group,BMD and BV/TV of diethylstilbestrol group,Qianggu Jiaonang group and each dose group of Echinops latifolius Tausch significantly increased,and the optical density of BMP-2 in diethylstilbestrol group,Qianggu Jiaonang group and Echinops latifolius Tausch medium dose group significantly increased(P<0.05 or P<0.01). Compared with the sham operation group,the body mass,Lee’s index during the model period and during the treatment period of the model group significantly increased,and the difference was statistically significant(P<0.05 or P<0.01).Compared with the model group,the Lee’s index of diethylstilbestrol group,Qianggu Jiaonang group and each dose group of Echinops latifolius Tausch during the treatment period significantly decreased,and the difference was statistically significant(P<0.05). Conclusion Mongolian medicine Echinops latifolius Tausch can increase the expression of BMD,BV/TV and BMP-2 in PMOP model rats and reduce Lee’s index,thereby effectively resisting PMOP.
