miR-150、c-Myb和TGF-β1在CNP睾丸生精功能异常中的作用 被引量:1

Roles of miR-150,c-Myb and TGF-β1 in abnormal testis spermatogenic caused by chronic nonbacterial prostatitis

摘要 目的:探讨miR-150、转录调节蛋白c-Myb和转化生长因子-β1(TGF-β1)在慢性非细菌性前列腺炎(CNP)睾丸生精功能异常中的表达水平,分析其影响机制及意义。方法:采用自身免疫反应诱导构建CNP动物模型,将前列腺注射生理盐水(对照组)和动物模型(实验组)的SD大鼠均置于智能培养气候箱中常规饲养至对数生长期,采用苏木精-伊红染色法检测两组前列腺腺泡结构。另根据实验组SD大鼠的睾丸活检情况分为生精功能正常组和生精功能异常组,比较生精功能正常组和生精功能异常组的hsa-miR-150、c-Myb、TGF-β1表达水平差异。通过ROC曲线分析hsa-miR-150、c-Myb、TGF-β1指标在CNP动物模型中生精功能受损情况。采用实时定量PCR(qRT-PCR)检测两组外周血中hsa-miR-150、c-Myb、TGF-β1表达水平差异。结果:实验组苏木精-伊红染色结果示前列腺腺泡腔被广泛破坏,间质及腺体内炎性细胞浸润明显,腺腔周围可见明显纤维结缔组织增生,与对照组比较(P<0.05),表明动物模型制作成功。实验组的hsa-miR-150、c-Myb、TGF-β1表达水平明显高于对照组,差异有统计学意义(P<0.05)。生精功能异常组的hsa-miR-150、c-Myb、TGF-β1表达水平明显高于生精功能正常组,差异有统计学意义(P<0.05)。结论:自身免疫反应诱导CNP动物模型可引起外周血hsa-miR-150水平呈高表达,进而激活下游靶向因子c-myb,诱导TGF-β1表达升高,其表达水平越高,预示着生精功能障碍越严重。 Objective:To study the expression and mechanism of miR-150,c-Myb and TGF-β1 in abnormal testis spermatogenic caused by chronic nonbacterial prostatitis(CNP).Method:CNP animal model was constructed by auto-immunogenicity.SD rats were divided into control group and studied group.Rats were routinely reared to logarithmic growth period in a smart culture climate chamber.Structure of prostate acinar was studied by HE staining.Rats were grouped as normal and abnormal based on the expression of testis spermatogenic within the studied group.The expression of hsa-miR-150,c-Myb and TGF-β1 in peripheral blood were determined by qRT-PCR.The damage of expression of hsa-miR-150,c-Myb and TGF-β1 were analyzed by ROC curve.Result:Compared with the control group,extensive destruction of the prostate acinar cavity is observed in the studied group by HE staining.Interstitial and glandular inflammatory cell infiltration is obvious.Obvious fibrous connective tissue hyperplasia is found around the glandular cavity.The expression of hsa-miR-150,c-Myb and TGF-β1 in studied group are much higher than those of control group(P<0.05).The expression of hsa-miR-150,c-Myb and TGF-β1 in the abnormal group are higher than those of the normal group(P<0.05).Conclusion:The expression of hsa-miR-150 in peripheral blood is higher in CNP animal model inducted by auto-immunogenicity which activates the downstream targeting factor c-Myb and induces the high expression of TGF-β1.The higher expression of hsa-miR-150 indicates more damage of the testis spermatogenic.
