黑木耳菌种常用培养基的筛选及菌丝体粗多糖抗氧化活性研究 被引量:4

摘要 为了降低黑木耳菌种公司制作菌种的生产成本,提高生产效率,缩短生产周期,提高黑木耳菌丝体的利用率,通过固体培养和摇瓶实验观察菌丝生长状况和时间,利用测量菌丝体干重的方法,从PDA培养基、PDA改良培养基、黄豆粉培养基和蛋白胨培养基4种工厂常用培养基中筛选出最优培养基,并从菌丝体中提取粗多糖,测定粗多糖对DPPH和羟基自由基的清除能力。结果表明,固体母种在PDA培养基中生长速度最快,在PDA改良培养基中质量最优;液体种子在PDA培养基中获得的菌丝干重最大。菌丝体中的粗多糖在PDA培养基中的提取率最高,粗蛋白含量在蛋白胨培养基中含量最高。菌丝体中粗多糖对DPPH和羟基自由基具有清除能力,并随浓度的增大而增大,具有较好的抗氧化作用。 In order to reduce the production cost,improve the production efficiency,shorten the production cycle and improve the utilization rate of Auricularia auricula mycelium,we selected the best medium from PDA medium,PDA improved medium,soybean powder medium and peptone medium,and extracted the crude polysaccharide from the mycelium to measure the scavenging ability of DPPH and hydroxyl radical free radicals.The results showed that the solid mother strain grew fastest in PDA medium,but the quality was the best in PDA improved medium,and the dry weight of hypha obtained from liquid seed in PDA medium was the largest.The crude polysaccharide in mycelia was extracted from PDA medium with the highest extraction rate and the crude protein content was the highest in peptone medium.Crude polysaccharides in mycelia can scavenge DPPH and hydroxyl radicals,and have good antioxidant activity with the increase of the concentration.
