皮肤鳞状细胞癌和基底细胞癌组织中VEGF、COX-2及ID-1的表达及临床意义 被引量:2

Expression and significance of VEGF,COX-2 and ID-1 in tissues of patients with cutaneous squamous cell carcinoma or basal cell carcinoma

摘要 目的研究皮肤鳞状细胞癌(SCC)和基底细胞癌(BCC)组织中血管内皮生长因子(VEGF)、环氧化酶-2(COX-2)及分化抑制因子-1(ID-1)的表达和临床意义。方法选取2016年3月—2019年3月重庆市江津区中心医院收治的皮肤恶性肿瘤患者76例为研究对象,其中SCC组和BCC组均38例。配对选取30例无恶性肿瘤的同期皮肤移植患者为对照组。采用免疫组织化学法检测VEGF、COX-2及ID-1的阳性表达率,RT-PCR检测mRNA的表达,Western blotting法检测蛋白相对表达量,并分析其相关性。结果 3组VEGF的阳性表达率、mRNA及蛋白表达的比较,差异有统计学意义(P<0.05);SCC组与BCC组高于对照组(P<0.05);SCC组高于BCC组(P<0.05)。VEGF、COX-2及ID-1的表达均呈正相关(P<0.05)。结论皮肤SCC与BCC的产生和进展与VEGF、COX-2及ID-1的高表达有关,且VEGF、COX-2及ID-1的表达均呈正相关。联合检测VEGF、COX-2及ID-1的表达对皮肤SCC与BCC的早期诊治具有重要意义。 Objective To study the expression levels and early diagnosis/treatment significances of vascular endothelial growth factor (VEGF),cyclooxygenase-2,Cyclooxygenase 2 (COX-2) and Inhibitor of DNA differentiation-1 (ID-1) in patients with squamous cell carcinoma of the skin (SCC) or basal cell carcinoma (BCC).Methods The study subjects were patients with skin malignant tumors who were admitted to our hospital from March 2016 to March 2019,including 38 patients in each group (the SCC group and the BCC group).Thirty patients with no malignant tumors at the same time were selected as the control group.The positive expression rate of each index was detected by immunohistochemistry,the mRNA expression of each index was detected by RT-PCR,the protein level of each index was detected by Western blotting method,and the correlation was analyzed.Results Statistically significant differences were found in VEGF positive expression rate,mRNA and protein expression among the three groups (P < 0.05);the comparison between each two groups showed that the VEGF indexes of SCC group and BCC group were higher than those of the control group (P < 0.05);the VEGF indexes of SCC group were higher than those of BCC group (P < 0.05).The expressions of VEGF,COX-2 and ID-1 were positively correlated (P < 0.05).Conclusion The production and progress of SCC and BCC are importantly related to the high expression of VEGF,COX-2 and ID-1,and the expressions of the three indicators have positive pairwise correlations.Combined detection of VEGF,COX-2 and ID-1 has important early diagnosis and treatment significance for SCC and BCC.
