摘要
人表皮生长因子受体2(human epidermal growth factor receptor 2,HER2)在多种癌症(如乳腺癌、卵巢癌)中高表达,是一个重要的肿瘤分子靶点,临床上迫切需要发展体内检测肿瘤组织HER2表达水平的分子影像学方法。核素分子影像即正电子发射断层成像(positron emission tomography,PET)/电子计算机断层扫描(computed tomography,CT)和单光子发射计算机断层成像(single-photon emission computed tomography,SPECT)/CT具有全身、半定量、实时和动态等内在优势,已经成为肿瘤精准诊断和治疗的重要手段,因此,核素标记HER2靶向的分子影像探针及其应用研究获得了快速发展。本文重点综述了核素标记HER2完整抗体和纳米抗体的分子影像探针及HER2阳性肿瘤PET/CT和SPECT/CT显像的研究进展。
Human epidermal growth factor receptor 2(HER2),as an important molecular target of tumor,is overexpressed in many kinds of cancers,such as breast cancer,ovarian cancer.It is urgent for the clinic to develop in vivo molecular imaging methods to detect HER2 expression in tumor tissues.Radionuclide-based molecular imaging,including positron emission tomography(PET)/computed tomography(CT)and single-photon emission computed tomography(SPECT)/CT,embracing whole body,semi-quantitative,real-time and dynamic inherent advantages,have become the key players in the precise diagnosis and treatment of tumors,therefore,radionuclide-labeled,HER2-targeted molecular image probes and the related applications have been developed extensively.In this paper,the research progress of molecular imaging probes based on radionuclide-labeled HER2 full-sized antibodies and nanobodies as well as their PET/CT and SPECT/CT imaging of HER2 positive tumors were concisely reviewed.
