有柄石韦对小鼠皮肤伤口愈合及转化生长因子-β1水平的影响 被引量:3

摘要 目的:观察有柄石韦对小鼠皮肤伤口愈合的作用及转化生长因子-β1(TGF-β1)水平的影响。方法:90只昆明小鼠均分为空白组、有柄石韦组及云南白药组,所有小鼠均于背中线做1 cm×1 cm的全层皮肤切除伤模型;除空白组外,其余2组均连续5 d局部给予相应药物,空白组给予生理盐水,每组随机选出10只小鼠测算伤口缩小比例,每组剩余的20只分别于给药完后第3、7、14及18天分4批次处死,采用HE染色观察创缘组织愈合情况,采用蛋白免疫印迹(Western blot)方法检测创缘组织的TGF-β1表达水平。结果:与空白组小鼠伤口比较,云南白药组及有柄石韦组在第3天、第7天、第14天及第18天时的伤口缩小比例较大;而与有柄石韦组比较,云南白药组在此时点的伤口缩小比例更高,差异有统计学意义(P<0.05);HE染色结果显示,有柄石韦组与云南白药组小鼠创缘组织肉芽组织较空白组的肉芽组织生长紧凑、无水肿现象,上皮化程度较高;Western blot结果显示,有柄石韦组和云南白药组创缘组织的TGF-β1表达量在第3及第7天持续增高、于第14天开始下降,空白组第3天含量最高、之后持续下降。结论:有柄石韦对皮肤伤口有一定促愈作用,其机制可能是刺激TGF-β1表达、加速伤口愈合。 Objective:To observe the effect of pyrrosiae petiolosa on wound healing in mice and the level of TGF-β1.Methods:90 mice with 1 cm×1 cm full-thickness skin excision models on their dorsal lines were equally divided into the control group,the pyrrosiae petiolosa group and the yunnanbaiyao group.The former group was given saline solution while the latter two groups were provided with relevant medicine for five consecutive days.Ten mice were randomly chosen from each group to measure wound contraction proportion while the remaining 20 mice were killed in batches on 3,7,14,and 18 d after administration.HE staining was employed to observe the healing of wound edge tissue and protein Western blot was used to check its TGF-β1 expression level.Results:Compared with the control group,the wound contraction proportion was larger in the pyrrosiae petiolosa group and the yunnanbaiyao group on 3,7,14,and 18 d.Meanwhile,yunnanbaiyao group witnessed higher degree of wound contraction than the pyrrosiae petiolosa group and the difference was statistically significant.HE staining result showed that granulation tissue of the wound edge in the pyrrosiae petiolosa group and the yunnanbaiyao group saw no edema and it revealed more compact growth and a higher level of epithelization.According to the Western blot result,TGF-β1 expression in the pyrrosiae petiolosa group and the yunnanbaiyao group continued to increase on 3 and 7 d,and started to decrease on 14 d.In the control group,it reached its peak on 3 d and continued to decline afterwards.Conclusion:Pyrrosiae petiolosa has some effect on wound healing.Its mechanism might be to stimulate TGF-β1 expression and therefore facilitate wound healing.
