摘要
在骨髓间充质干细胞(BMSC)的归巢、增殖、凋亡和成骨向分化等环节中,糖原合成酶激酶-3β(GSK-3β)为参与相关信号通路的重要蛋白。GSK-3β是一种多功能激酶,磷酸化的GSK-3β为其失活态。研究显示磷酸化的GSK-3β能促进细胞归巢、细胞增殖和成骨向分化。总体而言,负调控GSK-3β的活性水平,可促进BMSC骨生成,这已在多项体内、体外研究中通过锂离子或特异性抑制剂被证实。因此,负调控GSK-3β的活性水平可作为促骨修复的策略之一。本文主要对GSK-3β的基本性质、GSK-3β在BMSC的归巢、增殖、凋亡和成骨向分化等环节中发挥作用的主要信号通路,以及GSK-3β与相关通路关键蛋白的作用关系进行综述。
Glycogen Synthase Kinase‑3β(GSK‑3β)is a multifunctional kinase,which is an important component of diverse signaling pathways involved in the regulation of homing,proliferation,apoptosis and osteogenic differentiation of BMSC.GSK‑3βis inactive when it is phosphorylated.Phosphorylated GSK‑3βcan enhance the homing,proliferation and osteogenic differentiation of BMSC.In all,repression of GSK‑3βcan enhance bone formation of BMSC,which has been validated in a number of in vivo and in vitro studies by the application of lithium and specific inhibitors.This review aims to summarize recent studies on the properties of GSK-3β,the principal signaling pathways mediated by GSK-3βin related biological processes,and the relationship between GSK-3βand many other key proteins.
