Slug、IMP3和miR-221在食管胃交界腺癌的表达及其与E-cadherin的关系 被引量:5

摘要 目的探究Slug、胰岛素样生长因子ⅡmRNA结合蛋白3(insulin-like growth factorⅡmRNA-binding protein 3,IMP3)和微小RNA(microRNA,miR)-221在食管胃交界腺癌(adenocarcinoma of the esophagogastric junction,AEG)的表达及其与E-钙黏蛋白(E-cadherin)的关系。方法选取2017年12月至2019年6月邯郸市第一医院收集的AEG患者100例作为研究对象。收集同期体检者正常胃组织50例作为健康组。经过手术或全身PET-CT检测AEG转移情况。通过qPCR检测组织中miR-221的表达水平。通过免疫组化染色检测组织中E-cadherin、Slug、IMP3的表达。结果AEG组织中Slug、IMP3阳性率及miR-221相对表达量均显著高于健康组织(P<0.05)。性别、年龄及肿瘤大小对AEG组织中Slug、IMP3和miR-221的影响不大(P>0.05)。Ⅲ/Ⅳ分期、出现局部浸润、淋巴转移、远端转移患者的AEG组织中Slug、IMP3阳性率及miR-221显著升高(P<0.05)。且E-cadherin阳性组织中Slug、IMP3和miR-221显著低于E-cadherin阴性(P<0.05)。结论Slug、IMP3和miR-221在AEG中均过表达,与AEG转移有关,且与E-cadherin水平呈负相关,提示Slug、IMP3和miR-221可作为AEG的生物标志物。 Objective To investigate the expression of Slug,insulin-like growth factorⅡmRNA-binding protein 3(IMP3)and miR-221 in adenocarcinoma of the esophagogastric junction(AEG)and the relationship with E-cadherin.Methods A total of 100 AEG patients from Dec.2017 to Jun.2019 were selected as subjects.50 cases of normal gastric tissue were collected as healthy group.AEG metastasis was detected by surgery or whole body PET-CT.The expression level of miR-221 in tissues was examined by qPCR.The expression of E-cadherin,Slug and IMP3 in tissues was detected by immunohistochemical staining.Results The positive rates of Slug,IMP3 and miR-221 in AEG tissues were significantly higher than those in healthy tissues(P<0.05).Gender,age and tumor size had little effect on Slug,IMP3 and miR-221 in AEG tissues(P>0.05).The positive rate of Slug,IMP3 and miR-221 in AEG tissues of patients with stageⅢ/Ⅳstaging,local invasion,lymphatic metastasis and distal metastasis were significantly increased(P<0.05).And Slug,IMP3 and miR-221 in E-cadherin positive tissues were significantly lower than E-cadherin negative(P<0.05).Conclusion Slug,IMP3 and miR-221 are overexpressed in AEG,correlated with AEG metastasis,and negatively correlated with E-cadherin levels,suggesting that Slug,IMP3 and miR-221 can be used as biomarkers of AEG.
