五味子醇甲对小鼠抑郁行为的影响 被引量:4

Effects of Schisandrin on Depressive Behavior in Mice

摘要 目的探讨五味子醇甲干预后对血管性抑郁小鼠抑郁行为的影响。方法将40只小鼠随机分为0.9%氯化钠注射液组、模型组、阳性对照组和五味子醇甲组。模型组、阳性对照组和五味子醇甲组小鼠建立血管性抑郁模型,0.9%氯化钠注射液组进行假手术;手术后第8天0.9%氯化钠注射液组及模型组采用0.9%氯化钠注射液灌胃给药,阳性对照组采用20 mg/kg的盐酸度洛西汀灌胃给药,五味子醇甲组灌胃给药30 mg/kg的五味子醇甲。给药前及给药第7、15、28天时禁食8h后测量小鼠体重;给药第1、7、15及28天时对小鼠糖水偏好进行测定;在给药第1、15及28天时每只小鼠预先给予50 g鼠粮,24 h后称量剩余量确定小鼠摄食量;在给药第26天时行强迫游泳试验,在给药第27天行悬尾试验,在给药第28天行旷场试验。结果与模型组比较,给药第7、15及28天时阳性对照组及五味子醇甲体重显著增加(P<0.05),第15天及第28天阳性对照组及五味子醇甲组摄食量显著增加(P<0.05),第28天时阳性对照组及五味子醇甲组糖水偏好百分比显著升高(P<0.05);与模型组比较,采用五味子醇甲干预后可显著降低小鼠强迫游泳不动时间、悬尾不动时间、小鼠中央停留时间(P<0.05),并显著提高穿越格数、站立次数及修饰次数(P<0.05)。结论五味子醇甲对抑郁症小鼠进行干预后,可改善小鼠抑郁行为。 Objective To explore the effects of schisandrin on depressive behavior in mice with vascular depression. Methods A total of forty mice were randomly divided into a 0.9% sodium chloride injection group, a model group, a positive control group and a schisandrin a group. The model group, positive control group and schisandrin group established vascular depression model, and the 0.9% sodium chloride injection group received sham operation;on the 8 th day after operation, the model group was given 0.9% sodium chloride injection by gavage, the positive control group was given duloxetine hydrochloride by gavage, and the schisandrin a group was given 30 mg/kg schisandrin by gavage. The weight of mice was measured before administration and 8 hours after fasting on the 7 th, 15 th and 28 th days of administration;the preference of sugar water was determined on the 1 st, 7 th, 15 th and 28 th days of administration;on the 1 st, 15 th and 28 th days of administration, each mice was given 50 g rat food. After 24 hours, the surplus was weighed to determine the food intake;forced swimming test was performed on the 26 th day, tail suspension test on the 27 th day, and open field test on the 28 th day. Results Compared with the model group, the body weight of positive control group and schisandrin a group increased significantly on the 7 th, 15 th and 28 th day(P<0.05), the food intake of positive control group and schisandrin a group increased significantly on the 15 th and 28 th day(P<0.05), and the sugar and water preference percentage of positive control group and schisandrin a group increased significantly on the 28 th day(P<0.05). Compared with the model group, the intervention of schisandrin could significantly reduce the immobility time of forced swimming, the immobility time of tail suspension, the central residence time of mice(P<0.05), and significantly increase the crossing grid number, standing times and modification times(P<0.05). Conclusion Schisandrin a can improve the depressive behavior of depression mice after intervention.
