摘要
目的探讨T细胞免疫球蛋白黏蛋白分子3(TIM-3)与干扰素γ(IFN-γ)在肺腺癌的表达情况及其意义。方法以广州医科大学附属肿瘤医院的36例肺腺癌患者作为研究对象,收集患者肺组织及其外周血,密度梯度离心法分离外周血单核细胞,实时荧光定量PCR技术测定TIM-3与IFN-γ的mRNA相对表达量;免疫印迹和免疫组织化学技术测定蛋白表达量。结果肺腺癌患者癌组织TIM-3 mRNA与蛋白表达量均高于癌旁组织和正常组织(均P<0.05);伴有淋巴结转移肺腺癌患者的TIM-3呈高表达(P<0.05)。肺腺癌患者癌组织IFN-γmRNA与蛋白表达量均低于癌旁组织及正常组织(均P<0.05);在正常组织与癌旁组织中,伴有淋巴结转移肺腺癌患者的IFN-γ表达量高于未转移者(P<0.05)。癌组织中TIM-3、IFN-γ的mRNA与蛋白表达均呈负相关(r=-0.254,P=0.040;r=-0.312,P=0.010)。结论肺腺癌中TIM-3呈现相对高表达,且可能通过下调IFN-γ的表达而促进肺腺癌的生长和转移,这可能是肺腺癌治疗的潜在靶点之一。
Objective To investigate the expression and clinical significance of T cell immunoglobulin mucin 3(TIM-3)and interferonγ(IFN-γ)in lung adenocarcinoma.Methods Included in this study were 36 patients with lung adenocarcinoma patients from Cancer Hospital Affiliated to Guangzhou Medical University.The subjects were collected of lung tissues and peripheral blood samples.Peripheral blood mononuclear cells were isolated by density gradient centrifugation.Real-time fluorescence quantitative PCR was used to determine the relative expression of TIM-3 and IFN-γmRNA.Western blotting and immunohistochemical techniques were used to determine the protein expression of TIM-3 and IFN-γ.Results The mRNA and protein expression of TIM-3 was higher in cancerous tissues than that in paracancerous and normal tissues,of patients with lung adenocarcinoma(both P<0.05).Lung adenocarcinoma patients with lymph node metastasis had higher level of TIM-3 expression than those without(P<0.05).The expression levels of IFN-γmRNA and protein in cancerous tissues were lower than those in paracancerous and normal tissues,of patients with lung adenocarcinoma(both P<0.05).In normal and paracancerous tissues,the expression level of IFN-γwas higher in lung adenocarcinoma patients with with lymph node metastasis than those without(P<0.05).In cancerous tissues,TIM-3 was negatively correlated IFN-γin mRNA and protein expressions(r=-0.254,P=0.040;r=-0.312,P=0.010).Conclusion TIM-3 is relatively up-regulated in lung adenocarcinoma.TIM-3 possibly promotes the growth and metastasis of lung adenocarcinoma by down-regulating IFN-γexpression,which may be one of the potential targets for the treatment of lung adenocarcinoma.
