胚源性IL-1β与人类胚胎发育及种植的关系 被引量:2

Relationship between embryogenic IL-1βand human embryonic development and implantation

摘要 目的探讨胚胎培养液中胚胎分泌的白细胞介素1β(interleukin-1β,IL-1β)水平与胚胎发育及植入潜能的关系。方法选取首次行卵胞浆内单精子显微注射-胚胎移植(intracytoplasmic sperm injection-embryo transfer,ICSI-ET)手术的新鲜周期不孕患者80例,胚胎在体外采用单液滴单胚胎培养,在取卵受精后第3天进行胚胎移植,且移植胚胎数量均为2枚。收集新鲜周期移植胚胎相对应的卵裂期培养液微滴,采用酶联免疫吸附试验测定其上清的IL-1β水平。根据每周期胚胎种植率,将入组患者分A组43例(种植率为0%)、B组27例(种植率为50%)、C组10例(种植率为100%)。比较3组主要临床资料与胚胎实验室相关参数、在移植胚胎培养液中分泌的IL-1β水平、移植胚胎形态学累积评分;分析IL-1β水平、移植胚胎形态学累积评分之间的相关性。结果80例患者中临床妊娠37例,临床妊娠率为46.3%,其中单胎妊娠27例,双胎妊娠10例。3组主要临床资料与胚胎实验室相关参数差异均无统计学意义(P>0.05)。B组移植胚胎卵裂期培养液中IL-1β水平高于A组,C组高于A组和B组,差异有统计学意义(P<0.05)。3组移植胚胎形态学累积评分差异无统计学意义(P>0.05)。IL-1β水平与胚胎形态学累积评分呈正相关(r=0.587,P<0.05)。结论胚胎培养过程中胚胎分泌的内源性IL-1β与胚胎发育及植入存在密切相关性,其对胚胎的评估以及早期胚胎干预的研究可提供参考意义。 Objective To investigate the relationship between the level of interleukin-1β(IL-1β)secreted by embryos in embryo culture medium and embryo development and implantation potential.Methods Eighty patients with fresh cycle infertility who underwent the first intracytoplasmic sperm injection-embryo transfer(ICSI-ET)operation were selected.Embryos were cultured in vitro with single droplet single embryo.Embryo transfer was performed on the third day after oocyte retrieval and fertilization,and the number of transferred embryos was 2.The corresponding microdroplets of cleavage-stage culture fluid from fresh-cycle embryos were collected and the levels of IL-1βin their supernatants were measured by enzyme-linked immunosorbent assay.According to the implantation rate per cycle,the patients were divided into group A(43 cases,implantation rate was 0%),group B(27 cases,implantation rate was 50%),and group C(10 cases,implantation rate was 100%).The main clinical data of the three groups were compared with the relevant parameters of embryo laboratory,the level of IL-1βsecreted in embryo transfer medium,and the cumulative score of embryo transfer morphology.The correlation between the level of IL-1βand the cumulative score of embryo transfer morphology was analyzed.Results There were 37 clinical pregnancies in 80 patients,and the clinical pregnancy rate was 46.3%,including 27 singleton pregnancies and 10 twin pregnancies.There was no significant difference in the main clinical data and embryo laboratory parameters among three groups(P>0.05).The level of IL-1βin the culture medium of embryo cleavage stage in group B was higher than that in group A,and that in group C was higher than that in group A and group B,the difference was statistically significant(P<0.05).There was no significant difference in cumulative scores of morphology among the three groups(P>0.05).The level of IL-1βwas positively correlated with the cumulative score of embryo morphology(r=0.587,P<0.05).Conclusion Endogenous IL-1βsecreted by embryos during embryo culture is closely related to embryo development and implantation,and its evaluation of embryos and study on early embryo intervention can provide reference significance.
