摘要
目的研究LncRNA-ATB在胃癌细胞的表达及体外功能,为进一步筛选胃癌的有效诊治靶点奠定基础。方法设计LncRNA-ATB干扰序列,构建敲低LncRNA-ATB的胃癌细胞系并验证,进行克隆、增殖、迁移侵袭实验。结果LncRNA-ATB在胃癌细胞BGC-823、MGC-803中高表达,成功转染LncRNA-ATB干扰序列,并验证敲低效率,体外转染LncRNA-ATB干扰序列后,胃癌细胞克隆、增殖、迁移侵袭能力明显降低(P<0.05,P<0.01)。结论敲低LncRNA-ATB后胃癌细胞的克隆、增殖、迁移侵袭能力明显降低,表明其在胃癌进展中发挥重要的调控作用。
Objective To study the expression of LncRNA-ATB in gastric cancer cells and its function in vitro,laying a foundation for further screening of effective diagnosis and treatment targets of gastric cancer.Methods The LncRNA-ATB interference sequence was designed to construct a knockdown lncRNA-ATB gastric cancer cell line,which was then verified by cloning,proliferation,migration and invasion experiments.Results LncRNA-ATB was highly expressed in gastric cancer cells BGC-823 and MGC-803.The LncRNA-ATB interference sequence was successfully transfected,and the knockdown efficiency was verified.After in vitro transfection of the LncRNA-ATB interference sequence,the cloning,proliferation,migration and invasion ability of gastric cancer cells was significantly reduced(P<0.05,P<0.01).Conclusion After knockdown of LncRNA-ATB,the ability of cloning,proliferation,migration and invasion of gastric cancer cells was significantly reduced,which indicated that LncRNA-ATB played an important regulatory role in the progression of gastric cancer.
