摘要
目的探讨卵巢癌患者金黄色葡萄球菌L型感染与临床病理特征及鼠类肉瘤病毒癌基因(Kirsten rat sarcoma viral oncogene,KRAS)蛋白表达的关系。方法选取河南省肿瘤医院2016年1月-2019年6月收治的卵巢癌患者91例与卵巢良性肿瘤患者32例的临床资料,并对其存档于病理科的术后蜡块组织进行切片,采用革兰染色检测病灶组织中金黄色葡萄球菌L型感染情况,采用RT-PCR检测组织中KRAS mRNA相对表达量,采用免疫组化染色法检测病灶组织中KRAS蛋白表达情况,分析金黄色葡萄球菌L型感染与卵巢癌患者临床病理特征及KRAS蛋白表达的关系,金黄色葡萄球菌L型感染与KRAS蛋白阳性的相关性,以及金黄色葡萄球菌L型感染与KRAS mRNA和蛋白表达的关系。结果卵巢癌和卵巢良性肿瘤患者组织中金黄色葡萄球菌L型感染率分别为26.37%(24/91)和9.38%(3/32),癌组织中金黄色葡萄球菌L型感染率高于良性肿瘤组织(P=0.045);卵巢癌中KRAS mRNA相对表达量为(5.62±2.37)高于卵巢良性肿瘤中KRAS mRNA相对表达量(P<0.001);卵巢癌组织中KRAS蛋白表达阳性率为83.52%亦高于卵巢良性肿瘤组织(P<0.001)。金黄色葡萄球菌L型感染与卵巢癌的临床分期、组织学分级及腹腔淋巴结转移具有相关性(P<0.05);KRAS蛋白表达与临床分期、组织学分级具有相关性(P<0.05)。相关分析结果显示,卵巢癌中金黄色葡萄球菌L型感染与KRAS mRNA相对表达量呈现明显的正相关(r=0.478,P=0.021),卵巢良性肿瘤中,两者无相关性(r=0.178,P=0.134)。卵巢癌组织随着KRAS蛋白表达程度的增高,金黄色葡萄球菌L型感染率呈升高趋势(Z=7.036,P=0.008),而在卵巢良性肿瘤组织中金黄色葡萄球菌L型感染率与KRAS蛋白表达程度不存在线性趋势(P=0.148)。结论卵巢癌组织中金黄色葡萄球菌L型感染与患者临床病理分级、癌细胞转移有关,且与KRAS mRNA、蛋白表达存在线性趋势,可能与卵巢癌发生和进展有关。
OBJECTIVE To investigate the relationship between L-form infection of Staphylococcus aureus and clinicopathological features and expression of KRAS protein in patients with ovarian cancer.METHODS The clinical data of 91 patients with ovarian cancer and 32 patients with benign ovarian tumors admitted to Henan Cancer Hospital from Jan.2016 to Jun.2019 were selected,and the postoperative paraffin tissues achieved in the pathology department were sectioned.Gram staining was used to detect the L type infection of Staphylococcus aureus in the lesion tissue,RT-PCR assay was used to detect the relative expression of KRAS mRNA in tissues,the immunohistochemistry was used to detect the expression of KRAS protein in the focus tissue.The relationship between L-form infection of Staphylococcus aureus and clinicopathological features and KRAS protein expression in patients with ovarian cancer,the correlation between L-form infection of Staphylococcus aureus and KRAS protein positive,as well as the relationship between L-form infection of Staphylococcus aureus and KRAS mRNA and protein expression were analyzed.RESULTS The infection rate of L-form Staphylococcus aureus in the tissues of patients with ovarian cancer and benign ovarian tumor was 26.37%(24/91)and 9.37%(3/32),respectively.The infection rate of L-form Staphylococcus aureus in cancer tissues was significantly higher than that in benign tumor tissues(P=0.045).The relative expression of KRAS mRNA in ovarian cancer was(5.62±2.37),significently higher than that in benign ovarian tumors(P<0.001).The positive rate of KRAS protein expression in ovarian cancer tissue was 83.52%,which was also higher than that in benign ovarian tumor tissue(P<0.001).The L-form infection of Staphylococcus aureus was correlated with the clinical stage,histological grade and abdominal lymph node metastasis of ovarian cancer(P<0.05).The expression of KRAS protein was correlated with clinical stage and histological grade(P<0.05).The results of correlation analysis showed that there was a significant positive correlation between the L-form infection of Staphylococcus aureus and the relative expression of KRAS mRNA in ovarian cancer(r=0.478,P=0.021),and there was no correlation between the two in ovarian benign tumors(r=0.178,P=0.134).With the increase of KRAS protein expression in ovarian cancer tissues,the L-type infection rate of Staphylococcus aureus showed an increasing trend(Z=7.036,P=0.008),while there was no linear trend between l-type infection rate of S.aureus and KRAS protein expression in benign ovarian tumors(Z=2.092,P=0.148).CONCLUSION L-form infection of Staphylococcus aureus in ovarian cancer was related to the clinicopathological grade and cancer cell metastasis,and there was a linear trend with the expression of KRAS mRNA and protein,which may be related to the occurrence and progress of ovarian cancer.
