太行鸡胚PRLR基因表达与主翼羽和覆主翼羽长关系

Expression of PRLR gene and the feather length variation of primaries and secondary primaraies of Taihang chicken embryo

摘要 旨在探究催乳素受体(PRLR)基因不同剪切体表达对不同胚龄快慢羽太行鸡主翼羽和覆主翼羽生长影响。采集18和19胚龄太行鸡肝脏DNA、左翅皮肤组织RNA、右翅主翼羽和覆主翼羽,测量主翼羽和覆主翼羽长,通过PCR鉴定鸡胚羽型和性别、检测PRLR基因不同剪切体在鸡翅皮肤组织的表达变化,分析羽型、胚龄和性别对主翼羽和覆主翼羽长影响和PRLR剪切体表达变化,明确PRLR基因剪切体类型表达量与快慢羽鸡主翼羽和覆主翼羽长间关系。结果显示性别对主翼羽和覆主翼羽的长度均没有显著影响(P>0.05),但胚龄和羽型对两种羽毛长度均有显著影响(P<0.05)。18胚龄时快羽鸡主翼羽和覆主翼羽长显著长于慢羽鸡(P<0.05),19胚龄时快慢羽鸡间主翼羽长度仍然保持显著差异(P<0.05),而覆主翼羽的长度显著差异消失(P>0.05);18胚龄两种羽型鸡的主翼羽均长于覆主翼羽(P<0.05),而19胚龄慢羽鸡的主翼羽和覆主翼羽间长度差异消失(P>0.05)。鸡胚皮肤组织共发现PRLRS1和PRLRS2两种剪切体,胚龄和性别对二者的表达均没有显著影响(P>0.05),羽型影响PRLRS2的表达(P>0.05),19胚龄时慢羽PRLRS2表达显著高于快羽(P<0.05)。本试验推断快慢羽太行鸡18胚龄时主翼羽与覆主翼羽的长度差异,以及两种羽毛长度在快慢羽鸡间的差异已经凸现,但19胚龄慢羽鸡覆主翼羽生长速度增加,两种羽型鸡的覆主翼羽长度差异以及慢羽鸡两种羽毛间长度差异消失;慢羽鸡19胚龄PRLRS2高表达,可能促进了其覆主翼羽生长。该研究结果为揭示快慢羽鸡主、覆主翼羽长度差异的形成以及PRLRS2在调控羽型形成作用机制提供数据支持和理论参考。 To explore the effects of different spliceosomes of PRLR gene in Taihang chickens on the growth of the primary flight feathers and secondary primary flight feather at different embryonic ages.The liver DNA,the mRNA of primary flight feathers and secondary primary flight feather of Taihang chickens at 18(E18)and 19(E19)embryonic days were collected,and the length of the primary and the primary coverts were measured.The feather type and sex were identified by PCR,the expression changes of different spliceosomes of PRLR gene were semi-quantified,and the effects of feather type,embryo age and sex on the primary and the primary covert length and PRLR spliceosomes expression were analyzed.Results showed that sexes had no significant effects on the length of primary and the primary coverts(P>0.05),while ages and feather types both influenced them(P<0.05).The length of the two type feathers of early-feathering chickens at 18 E were both longer than the late-feathering chickens(P<0.05).And the length difference of primary flight feather between the two feather types of chickens was also significant at E19(P<0.05),but that of the secondary primary flight feather disappeared(P>0.05).The length of primary feather was significantly longer than the secondary flight feather(P<0.05)in the two feather types of chickens at E18,but the difference of slow-feathering chickens disappeared at E19(P>0.05).Two kinds of spliceosomes of PRLR were expressed in chicken skin,but ages and sexes had no effects on the expression of the two spliceosomes(P>0.05).While feather types only effected the expression of PRLRS2(P<0.05),showing that the expression of PRLRS2 was significantly higher in slowfeathering chickens at E19.The length difference of primary flight feathers and secondary primary flight feathers was shown in the two feather types of Taihang chickens at 18 embryonic day.However the length difference of secondary primary flight feather disappeared at E19.The higher expression of the PRLRS2 in the slow feathers chickens at 19 embryonic day might promote the growth of the secondary primary flight feathers.These results provided data for the regulating role of PRLRS2 in the growth of secondary primary flight feathers of slow-feathering chickens,and a reference for the formation of feather type.
