摘要
目的分析泛醌蛋白1(UBQLN1)及上皮-间质转化(EMT)标志分子在食管鳞癌(ESCC)组织中的表达及其相关性,探讨UBQLN1对ESCC细胞迁移和侵袭的影响。方法RT-qPCR和western blot检测ESCC癌组织及对应癌旁组织中UBQLN1的mRNA和蛋白表达;RT-qPCR检测ESCC癌组织及对应癌旁组织中EMT标志分子(E-cadherin、N-cadherin、Vimentin)的mRNA表达;构建UBQLN1 siRNA和过表达重组质粒,分别转染ESCC细胞系EC9706、HET-1A和TE-1;采用划痕试验和Transwell试验检测各细胞的迁移和侵袭能力;RT-qPCR和western blot检测各组EC9706细胞中EMT标志分子的mRNA和蛋白表达。结果ESCC癌组织中UBQLN1的mRNA和蛋白表达均高于癌旁组织(均P<0.05);ESCC癌组织中E-cadherin mRNA的表达低于癌旁组织(P<0.05),而N-cadherin和Vimentin mRNA的表达均高于癌旁组织(均P<0.05);ESCC组织中UBQLN1与E-cadherin的mRNA表达呈负相关(r=-0.5173,P<0.001),而与N-cadherin和Vimentin的mRNA表达呈正相关(r=0.7803、0.6856,P<0.001)。下调UBQLN1表达增加EC9706、HET-1A和TE-1细胞中E-cadherin的表达,降低N-cadherin和Vimentin的表达,抑制其迁移和侵袭,而过表达UBQLN1下调E-cadherin的表达,上调N-cadherin和Vimentin的表达,促进细胞迁移和侵袭。结论UBQLN1能诱导EMT,促进ESCC细胞的迁移和侵袭。
Objective To determine the expression of UBQLN1 and epithelial-mesenchymal transition(EMT)marker molecules and their correlation in oesophageal squamous cell carcinoma(ESCC),and to study the effects of UBQLN1 on migration and invasion of ESCC.Methods The expression levels of UBQLN1 mRNA and protein were determined in ESCC and para-carcinoma tissues by RT-PCR and Western blot,respectively.EMT marker molecules E-cadherin,N-cadherin and Vimentin were detected by RT-PCR.UBQLN1 siRNA and overexpression recombinant plasmids were constructed and transfected into EC9706,HET-1A and TE-1 cells.Wound healing assay and transwell assay were performed to determine the migration and invasion of cells.RT-PCR and Western blot were used to determine the expression of EMT-related molecules in EC9706 cells.Results Compared with para-carcinoma tissues,the expression of UBQLN1,N-cadherin and Vimentin increased but the expression of E-cadherin decreased in ESCC(P<0.05).UBQLN1 expression was negatively correlated with E-cadherin expression(r=-0.5173)but positively related to expression of N-cadherin(r=0.7803)and Vimentin(r=0.6856)in ESCC(P<0.001).The down-regulation of UBQLN1 expression enhanced E-cadherin expression,reduced N-cadherin and Vimentin expression and inhibited migration and invasion in EC9706,HET-1A and TE-1 cells.Conversely,UBQLN1 overexpression suppressed E-cadherin expression,up-regulated N-cadherin and Vimentin expression and promoted migration and invasion in EC9706,HET-1A and TE-1 cells.Conclusion UBQLN1 induces EMT and promotes migration and invasion in ESCC.
