七叶皂苷钠对骨骼肌缺血再灌注的影响 被引量:5

Effect of precondition with sodium aescinate on ischemia-reperfusion injury of skeletal muscle

摘要 [目的]探讨七叶皂苷钠对大鼠骨骼肌缺血再灌注损伤的影响。[方法] 30只SD大鼠分随机分为三组,每组10例。七叶皂组大鼠于尾静脉注射七叶皂苷钠溶液(1 ml),安慰剂组大鼠注射等量0.9%生理盐水,无创动脉夹闭右侧股动脉4 h,再灌注8 h。正常组给予药物,亦未行抽血再灌注处理。取静脉血、鼠右下肢骨骼肌组织进行检测。[结果]再灌注后的第8 h时,七叶皂组血液MB、LPC及LPO水平显著低于安慰剂组(P<0.05);七叶皂组iNOS表达低于安慰剂组,而GPX1、CAT及SOD2显著高于安慰剂组(P<0.05);七叶皂组IL-1、IL-6及TNF-α显著低于安慰剂组(P<0.05)。组织形态方面,安慰剂组肌纤维紊乱并水肿、炎性细胞浸润;而七叶皂组水肿与炎性细胞浸润显著减轻。[结论]七叶皂苷钠可减轻骨骼肌缺血再灌注损伤。 [Objective] To explore the effect of precondition with sodium aescinate on ischemia-reperfusion injury of skeletal muscle in rats. [Methods] A total of 30 SD rats were randomly divided into three groups with 10 animals in each group. The animal in the aescinate group were given 1 ml of sodium aescinate intravenously, whereas those in placebo group received equal volume of normal saline, then were atraumatically clipped right femoral artery for 4 hours, and re-perfused for another 8 hours.By contrast, the rats in the normal control group had no drug and ischemia-reperfusion treated. The venous blood and skeletal muscle samples were harvested for assessments at 8 hours after reperfusion. [Results] At 8 hours after reperfusion, the aescinate group had significantly lower levels of blood MB, LPC and LPO(P<0.05);significantly lower expression of iNOS, whereas significantly higher expression of GPX1, CAT and SOD2(P<0.05);in addition, significantly lower blood levels of IL-1, IL-6 and TNF-α than the placebo group(P<0.05). With regard to histological observation, remarkable edema and infiltration of inflammatory cells were seen in the placebo group, whereas these pathological changes significantly relieved in the aescinate group.[Conclusion] The sodium aescinate does have protective role against ischemia-reperfusion injury to skeletal muscle.
