平喘颗粒对哮喘小鼠肺组织Eotaxin和CCR3表达的影响 被引量:6

摘要 目的观察平喘颗粒对哮喘小鼠肺组织Eotaxin和CCR3表达的影响。方法30只BALB/c小鼠随机分对照组、哮喘模型组、哮喘+平喘颗粒组,每组10只。哮喘模型组、哮喘+平喘颗粒组腹腔注射及雾化吸入卵清蛋白(OVA)造成慢性哮喘动物模型,对照组腹腔注射及雾化吸入等体积生理盐水代替OVA激发;从第1次雾化激发的当天开始,哮喘+平喘颗粒组每天给予中药药液灌胃,持续给药8周,激发阶段则每次激发前1 h灌胃;对照组和哮喘模型组给予等体积的生理盐水灌胃,灌胃时间同哮喘+平喘颗粒组。检测各组小鼠肺泡灌洗液(BALF)中嗜酸性粒细胞(EOS)的数量,采用蛋白质印迹法检测肺组织Eotaxin和CCR3蛋白表达水平。结果与对照组比较,哮喘模型组和哮喘+平喘颗粒组BALF中EOS计数增多(P<0.05),且肺组织Eotaxin和CCR3蛋白的表达水平增高(P<0.05)。与哮喘模型组比较,哮喘+平喘颗粒组BALF中EOS计数和肺组织中Eotaxin、CCR3蛋白的表达均降低(P<0.05)。结论平喘颗粒可以通过降低哮喘小鼠肺组织中Eotaxin、CCR3蛋白的表达,抑制EOS向气道的聚集浸润,从而缓解哮喘气道的炎症反应。 Objective:To observe the effect of Pingchuan Granules on the expression of Eotaxin and CCR3 in lung tissue of asthmatic mice.Methods:30 BALB/c mice were randomly divided into 3 groups:control group(group A),asthma model group(group B),asthma+Pingchuan Granules group(group C),10 mice in each group.In group B and group C,intraperitoneal injection and nebulized inhalation of ovalbumin(OVA)were applied to establish chronic asthma animal models.In group A,intraperitoneal injection and nebulized inhalation of equal volumes of saline were used instead of OVA challenge.From the day of the first atomization challenge,group C was given intragastric administration of Chinese medicine liquid for 8 consecutive weeks.In the challenge phase,the Chinese medicine liquid was given intragastric administration 1 h before each challenge.Group A and group B were given intragastric saline at the same volume for the same time as group C.The numbers of eosinophils(EOS)in bronchoalveolar lavage fluid(BALF)of each group of mice were calculated.The expression levels of Eotaxin and CCR3 proteins in lung tissue were detected by Western blot.Results:Compared with group A,the number of EOS in BALF of group B and group C were increased(P<0.05)and the expression levels of Eotaxin and CCR3 proteins in lung tissue were increased(P<0.05).Compared with group B,the number of EOS in BALF and the expression levels of Eotaxin and CCR3 proteins in lung tissues in group C were all reduced(P<0.05).Conclusion:Pingchuan Granules can alleviate the inflammatory response of asthma airway by reducing the expression levels of Eotaxin and CCR3 proteins in the lung tissue of asthmatic mice and inhibiting the accumulation and infiltration of EOS into the airway.
