摘要
目的对慢性阻塞性肺疾病(COPD)大鼠模型给予化痰活血降气方干预,探究其在COPD中的抗炎机制。方法随机将36只SPF级wistar大鼠分为空白组,模型组,阳性对照组,化痰活血降气方大、中、小剂量组。采用气管内滴注脂多糖(LPS)加烟熏法复制COPD模型,造模成功后治疗组中药灌胃,阳性对照组醋酸泼尼松片灌胃。观察大鼠肺组织病理学变化;检测大鼠肺脏、脾脏、胸腺指数;检测大鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、内皮素-1(ET-1)的水平。结果模型组肺组织病理改变较空白组明显加重,化痰活血降气方大、中剂量组可明显减轻肺组织病理改变。与空白组比较,模型组肺脏指数显著增高(P<0.01),脾脏指数显著降低(P<0.05);与模型组比较,化痰活血降气方大剂量组肺脏指数显著降低(P<0.01);与阳性对照组比较,大剂量组肺脏指数无统计学差异(P>0.05)。与空白组比较,模型组大鼠血清TNF-α、IL-1β、ET-1水平显著增高(P<0.01);与模型组比较,化痰活血降气方各剂量大鼠血清TNF-α、IL-1β、ET-1水平显著降低(P<0.05或P<0.01);与阳性对照组比较,大剂量组血清TNF-α、ET-1水平无统计学差异(P>0.05)。结论化痰活血降气方可通过降低大鼠血清TNF-α、IL-1β、ET-1等炎症因子水平缓解COPD的炎症反应,降低肺脏指数,对COPD起到治疗作用。
Objective:To explore the anti-inflammatory mechanism of Huatan Huoxue Jiangqi Decoction in rats model of chronic obstructive pulmonary disease(COPD).Methods:36 SPF wistar rats were randomly divided into the blank group,model group,positive control group,large-,medium-and small-dose groups of Huatan Huoxue Jiangqi Decoction.The COPD model was replicated by intratracheal infusion of lipopolysaccharide(LPS)and fumigation.After successful modeling,the treatment group was gavage with traditional Chinese medicine,and the positive control group was gavage with prednisone acetate tablets.The pathological changes of rats′lung tissues were observed;the lung,spleen and thymus index of rats was detected;the levels of tumor necrosis factor(TNF)-α,interleukin(IL)-1βand endothelin(ET)-1 in the serum of rats were detected.Results:The pathological changes of lung tissue in the model group were significantly worse than those in the blank group.The large-and mediumdose groups of Huatan Huoxue Jiangqi Decoction could significantly reduce the pathological changes of lung tissue.Compared with the blank group,the lung index of the model group was significantly higher(P<0.01),and the spleen index was significantly lower(P<0.05);compared with the model group,the lung index of the large-dose group of Huatan Huoxue Jiangqi Decoction was significantly lower(P<0.01);compared with the positive control group,there was no statistical difference with the lung index in the large-dose group(P>0.05).Compared with the blank group,the serum levels of TNF-α,IL-1β,and ET-1 in the model group were significantly increased(P<0.01);compared with the model group,the levels of serum TNF-α,IL-1βand ET-1 were significantly reduced(P<0.05 or P<0.01);compared with the positive control group,there was no significant difference in the levels of serum TNF-αand ET-1 in the large-dose group(P>0.05).Conclusion:Huatan Huoxue Jiangqi Decoction can alleviate the inflammatory response of COPD by reducing the level of inflammatory factors such as TNF-α,IL-1β,ET-1 in the serum of rats,and reduce the lung index,which has a therapeutic effect on COPD.
