刺猬皮提取工艺优化及其提取物TLC鉴别研究 被引量:1

摘要 目的优选出刺猬皮的最佳工艺参数并建立其提取物的TLC法。方法以总氨基酸含量和干膏得率为指标,通过单因素试验,结合正交试验-综合评分法考察乙醇浓度,料液比,提取时间及提取次数对提取工艺影响;同时以谷氨酸,甘氨酸,天门冬氨酸为鉴别指标,对刺猬皮提取物进行TLC鉴别。结果最佳提取工艺为:取刺猬皮炭品粉末,料液比为1∶14,70%乙醇提取120 min,提取1次;该提取物的TLC鉴别斑点清晰,分离度较好。结论该实验优选的提取工艺参数稳定、可靠,可为刺猬皮制剂制备提供实验参考。 Objective To optimize the technological parameters of Corium erinacei and establish TLC for its extract. Methods By using total amino acid content and dry paste yield as indexes,the effects of ethanol concentration,solid-liquid ratio,extraction time and extraction times on extraction process were investigated by single factor test and orthogonal test-comprehensive scoring method. Meanwhile,glutamic acid,glycine and aspartic acid were used as identification indexes to identify the extracts from Corium erinacei by TLC. Results The best extraction process was as follows: the charcoal powder of Corium erinacei was extracted by70% ethanol for 120 min with the ratio of material to liquid being 1: 14. The TLC of the extract showed clear spots and good separation. Conclusion The optimized extraction parameters are stable and reliable. This method can provide experimental reference for preparation of Corium erinacei.
